Abstract:目的将铁离子导入到大鼠的感觉运动皮质内,造成大鼠的外伤后癫痫的动物模型,观察大鼠术后癫痫发作的行为学改变。方法用离子导人法将铁离子导人到SD大鼠大脑皮质内,通电时间为10min,通电电流为200μA。对照组大鼠给予相同的手术操作,但不导入铁离子。结果实验组内20只大鼠有18只出现癫痫发作,癫痫模型制作成功率为90%;对照组内20只大鼠内有一只出现癫痫发作,癫痫模型制作成功率为5%。结论用离子导人法制作大鼠外伤后癫痫动物模型的通电电流以200μA为最佳条件,通电时间为10min。制造出来的模型的成功率较高,且为急性模型。

Abstract:Objective To find out a method of constructing a Neisseria gonorrhoeae genital infection mouse model.Method In our experiments,ICR mice were injected Estradiol(E2) and inplanted Estriol(E3) pellet respectively,then challenged with Neisseria gonorrhoeae WHO-A in the female mice vagina.The genital swab were verified by T-M culture media,the vaginal excretes smears were stained to determine different effect of the hormone on mouse Neisseria gonorrhoeae infection.Result Compared with non-estrogen-treated mice,estradiol(E2) treated mice have Neisseria gonorrhoeae short-lived in the vagina.And there was no gonococci in estriol(E3) treated mice like control group.Conclusion Estrogen,especially estradiol(E2) has the effect on promoting Neisseria gonorrhoeae vaginal colonization in mice.

Abstract:Objective To study the specific killing effect of recombinant expression vector containing human breast cancer DF3 promoter and diphtheria toxin A fragment on human breast cancer cells in vivo.Methods thirty-six BALB/C nude mice were divided into four groups at random,each group has nine animals.Human breast cancer cells of DF3 positive and negative(MCF-7 and MDA-MB-231)were inoculated into corresponding animals respectively.Recombinant expression vector PGL3-DF3-DTA and PGL3-DF3 were inoculated into corresponding tumor when the animals grew up tumor.The tumor weight were measured and calculated continually.We killed the animals in different time and observed the morphological changes by means of light microscope.The apoptosis index of cells was evaluated by in-situ TUNEL.Immunocytochemical methods were used to detect the condition of lymphanglogenesis and angiogenesis in tumor.Results Human breast cancer xenograft model was established in nude mice successfully.The human breast cancer cells treated by PGL3-DF3-DTA showed a typical apoptosis morphology and TUNEL detection analysis revealed the apoptosis index of the treatment groups was significantly enhanced,which was associated with time and dosage.The expression of LYVE-1 and F8 were down regulated.Conclusion Recombinant expression vector PGL3-DF3-DTA could produce specific killing effect on human breast cancer cell line of DF3 positive in vivo.

Abstract:Objective To study the regulation of BV antibody level of captive-bred cynomologus monkeys.Method BV antibodies of 409 captive-bred cynomolgus monkeys selected randomly were monitored strictly with BV ELISA.Results Nearly all the newborn monkeys had BV antibody,the BV antibody level changes with age increase.At fifth-month old,the BV antibody positive rate reaches the lowest(13.1%) level.Conclusions the regulation of BV antibody level of captive-bred cynomolgus monkeys can be characteristic as having high level after born and decreasing with its age increase as well as increasing after fifth-months old,it is the optimal time for weaning at fifth-month old so as to obtain more BV antibody negative monkeys.

Abstract:Objective The effect factors on blood glucose of KK/Upj-A^y/J mice with nutrition, age and sex. Methods Tail blood with experimental group mice and comparative group mice were measured by blood glucose detector. Results Either blood glucose level or ratio to all mice, experimental group were significantly highter than that of comparative group. On comparative group, blood glucose level is low on 8 months old. On the other hand, experimental group mice in both sex, age have a few bearing on blood glucose level. The sex has almost not an influence on all animal experiment. Conclusion I .nutrition components is main factors which affect blood glucose at same genetic background with A^ygene; 2.Another effect factor is the age of the mice; 3.The sex is not an important factor to blood glucose in the KK/Upj-A^y/J mice.

Abstract:Objective To observe the effects of iodine excess on expressions of neuron specific enolase(NSE) and glial fibrillary acidic protein(GFAP) in filial generation of rats.Methods The Wistar rats were divided into five groups by randomized design.The rats were drinking water at different iodine levels(NI,5HI,10HI,50HI and100HI).After being fed three months,the rats were mated and their filial generations at the age of 60 day observed.The serum thyroid hormones at the age of 60 day were measured.Immunohistochemical method combining with image analysis techniques were used to demonstrated expressions of NSE and GFAP in hippocampus of both control and iodine excess rats.Results The results of serum thyroid hormones revealed the decreasing tendency in experimental groups by gradually increasing iodine intake,however,hypothyroidism were obviously in 100HI group.Immunohistochemical staining and morphological images analysis of NSE positive neurons in hippocampus CA3 area indicated that numerical density on area(N-A),volume density(V-V) and the average of grey scale were gradually decreased coincidenced with iodine excess levels,furthermore,statistically significance in 100HI group.GFAP positive astrocytes in hippocampus CA3 area showed that the changes of the astrocytes were insignificant.Conclusions NSE expressions were decreased obviously in 100HI group at the age of 60 day.Iodine excess can cause brain under development in their filial generation,and the pathogenesis maybe related to hypothyroidism induced by iodine excess.

Abstract:目的建立适用于Lansendorff离体心脏灌流大鼠心肌梗死的动物模型,为评价干细胞移植对急性心肌梗死后的心功能变化提供基础。方法选用Sprague-Dawley(SD)大鼠16只,结扎其左冠状动脉前降支中远1/3处,在结扎前后通过MPA多导生理记录仪连续描记心电图;4周后再次开胸进行Langendorff离体心脏灌流测定左室心功能和心肌组织病理学检查;另选仅开关胸后存活的10只SD大鼠作为对照组。结果造模成功率为62.50%(10/16);心电图动态监测在冠脉结扎后出现ST-T抬高的融合波,30min后可见病理性Q波;4周后Langendorff离体心脏灌流装置系统检测显示左室收缩压峰值(LVSP)、左室内压等容相最大上升及下降速率(+dp/dtmax,-dp/dtmax)等指标较对照组降低,左室舒张末压峰值(LVEDP)则反之;病理组织切片可见结扎区域心肌纤维排列紊乱、坏死心肌被纤维组织取代。结论通过结扎左冠脉前降支的方法,4周后能够形成稳定的适用于Langendorff离体心脏灌流的心肌梗死动物模型,该模型能应用于干细胞移植对心脏功能影响的研究。

Abstract:Objective To study the protective effect and the mechanism of anti-myocardial-ischemia-reperfusion injury of Shen-fu injection through ligating the left anterior descending(LAD) of rats in vivo.Methods The 46 rats were divided into 5 groups randomLy:(1) no-operation group;(2) the control group 1: 30-minute ischemia;(3) pretreatment group 1 with Shen-fu: 30-minute ischemia;(4) the control group 2: 30-minute ischemia followed by 10-minute reperfusion;(5) pretreatment group 2 with Shen-fu: 30-minute ischemia followed by 10-minute reperfusion.The content of malondialdehyde(MDA) and the expression of antioxidant genes of superoxide dismutase 1(SOD1) gene and glutathione S-transferase gene were measured,the ultrastructure of the myocardial cells by transmission electron microscope was observed.The mitochondrial stereology analysis was done.(Results The) MDA of the pretreatment groups with Shen-fu was powful decreased compare with the control groups.The significant decreasing the lesion of the myocardial cell and mitochondrial in ultrastructure were found in the pretreatment groups with Shen-fu.The stereology analysis display that the mitochondrial in the control groups have significant changed compare with the no-operation group.And the mitochondrial in the pretreatment group with Shen-fu was a little different from the no-operation group.Shen-fu up-regulated the expression of SOD1 gene and glutathione S-transferase gene.Conclusion The Shen-fu protect the myocardial cell by reducing the lipid peroxidation.Their protective effects may be partly related to up-regulating the antioxidant gene that increase the ability of antioxidant.

Abstract:Objective To observe effect of Levofloxacin gel against Mycobacterium tuberculosis in vitro and safty of bronchial intervention therapy. Methods The MIC and MBC of Levofloxaein and Levofloxacin gel were measured by handwork and instrument methods, the safety of Levofloxacin gel was assessed by bronchial intervention therapy using rabbits as experiment animals. Results The MIC of Levofloxacin gel to M.tubereulo H37Rv, M.boris and M.phlei was 0.5,0.5 and 1.0 mg/L; the MBC of that was 1.0 ,1 .0 and 1.5 mg/L respectively. The MIC and MBC of Levofloxacin gel and Levofloxacin had no significant difference without any harmful effect. Conclusion the effect of Levofloxacin gel and Levofloxacin against Mycobacterium tuberculosis was equivalent, carbomer in the formulation did not affect the activity of Levofloxacin against Mycobacterium tuberculosis; meanwhile Levofloxacin gel which contains carbomer was safe in bronchial intervention therapy.

Abstract:Objective To study the defend function of lyophilized bovine colostrums on alcohol liver injury in mice.Methods Objective Lyophilized Bovine Colostrums(350 mg/kg) 3 d.The alcohol model group also used 50% alcohol(12 mL/kg) together.The control group only used distilled water.The experiment tested biochemistry and alcohol liver injury in adipose.Results The Lyophilized Bovine Colostrums group and alcohol model group GSH,HDL-C increased significant and deceased The content of MDA,Its protect effect was also proved by liver patholgical changes in total score.Conclusion Lyophilized Bovine Colostrums had protectived effect on alcohol liver injury.

Abstract:Objective To detect the function of Tiaoneixiaobiwan in enhancing the angiectasia of the capillaries and improving the microcirculation in mice's ala auris,and thus to explore the effects of Tiaoneixiaobiwan on the microcirculation of the mice's ala auris.Methods The method we used here was administering the drug directly.Results Tiaoneixiaobiwan could enhance the angiectasia of the capillaries and improve the microcirculation in mice's ala auris.(P<0.05,P<0.01,P<0.0001).Conclusion The fact that Tiaoneixiaobiwan could enhance the angiectasia of the capillaries and improve the microcirculation has provided the evidences for its application in clinical medicine.

Abstract:Objective To explore the feasibility of bareneck goby as a laboratory animal. Methods The natural distribution aquatic environment investigation, the biology parameter measurement, the gonad tissue slice observation and the artificial multiplication experiment were carried out. Results With characteristics of small body, omnivorousness, quick increase and strong reproductive capacity, bareneck goby is advantageous for raise and management in the laboratory. Concolusion goby is suitable to be used as aquatic laboratory animals.

Abstract:The use of experimental animals, mostly rodents, in biomedical research and especially in oncology and immunology should be acknowledged with respect, recognizing the contribution of animal experimentation in the fascinating scientific progress in these disciplines of research. It is an obligation of the investigator to justify the scientific and ethical aspects of each study requiring the use of animals. The international guiding principles for using animals in biomedical research are well defined and have been distributed worldwide by the International Council for Laboratory Animal Science (ICLAS) since 1956, when this Organization was founded. In Poland the ICLAS philosophy and principles are highly respected and were implemented firstly by the members of the Commission on Biology of Experimental Animals appointed in 1962 by the Department of Medical Science of the Polish Academy of Science in Warsaw. Animal Protection Acts, first proclaimed in 1928 were gradually modified and improved. Actual legislation (enacted in 1997, 2003 and 2005) is consistent with EU Directives (86/609/EEC) and follows the internationally recommended principles that include ICLAS guidelines concerning animal welfare and care condition in biomedical research. The problem of "alternative methods" is briefly discussed.

Abstract:Objective To develop a fast and easy-to-use kit based on GICA(gold immunochromatography assay) for the detection of human Influenza-A virus.Method Monoclonal antibody recognizing the NP of Influenza-A was conjugated with 40nm colloidal gold particle to prepare a kit.The conjugate reacts with Influenza-A antigens in the sample and migrates forward to the nitrocellose,on which another monoclonal antibody was pre-immobilized,thus producing noticeable band by naked eyes.Results All 16 strains of Influenza-A subtype virus detected by GICA show positive reaction.37 samples of health people show negative reaction.Three strains were used to evaluate the sensitivity of GICA.The kit has the same sensitivity as the production of Quidel inc.Conclusion GICA is a sensitive,specific and fast assay for the detection of Influenza-A virus.

Abstract:Toxicity pathology is the footstone of medicine preclinical safety evaluation. In practice, many problems were exist in pathological examination such as criterion were discordance, operating procedure was nonstandard and labanimal pathology study was unsubstantially, which severely threaten the running and effect of GLP. Therefore, toxicity pathology laboratory should execute the standardization of examination and promote laboratory certification and accreditation. It also was very important that strengthening research of comparative histology and comparative pathology in labanimal and building toxicity pathology database. All of this was to fundamentally accelerate the level of medicine safety evaluation.

Abstract:Non-human primates, which are extensively used in biomedical researches, play an important role in the study of physiology, pathology, virology, pharmacokinetic, toxicology, reproduction physiology, immunology, genetics, gene engineering drug and biological medicine and so on. Therefore, the animal welfares should be emphasized in research works. According to our practices and demand of animal welfares, this paper summarized that the veterinarian and animal nursing act in the welfares of non-human primates, which includes disease monitor, diagnosis, therapy, prevention, experimental operation, catching, securing, composing, anaesthesia, odynolysis and euthanasia etc.
