    • Influence of Cartilage Polysaccharide on Expression of Bcl-2 and Bad Protein in S180 Tumor-bearing Mice

      Abstract:目的通过软骨多糖对S180荷瘤小鼠实体瘤Bcl-2和Bad蛋白表达的影响研究软骨多糖抑瘤作用机理。方法皮下注射S180细胞建立动物实体瘤模型,将小鼠随机分为生理盐水对照组和软骨多糖给药组,连续腹腔注射生理盐水或软骨多糖21 d,取实体瘤制成石蜡切片;应用常规HE染色法观察细胞形态学变化,免疫组化SP法检测Bcl-2和Bad的表达情况,并比较酶解法和加热法进行的抗原修复对脱片率的影响。结果软骨多糖有效抑制了S180肉瘤的生长,Bcl-2的阳性表达率由(70.45±8.89)%降至(24.19±4.32)%,Bad的表达率由(4.14±1.35)%升至(18.89±3.15)%。酶解法进行抗原修复的脱片率要小于加热法。结论软骨多糖通过显著降低Bcl-2的表达水平和提高Bad的表达水平而诱导细胞凋亡,从而抑制了S180肿瘤细胞的生长;软骨多糖是一种新型的抗肿瘤活性物质。

    • RAPD Analysis of Murine Hepatoma 22 Clonal Cell Strains

      Abstract:Objective To detect the genetic stability in four elonal cell strains of murine hepatoma 22 with RAPD analysis. Methods Genomic DNAs from H22-H8D8,H22-H2G4,H22-H2E10,and H22-H2C4 were amplified by RAPD with 47 10bp random primers. Results The most amplified products were monophic, 4 of the 47primers generated polymorphic profile in four clonal cell strains. A muhiband profile were observed for all the primers with the totle number of bands per primer varying from 4 to 10 mostly, and these bands ranged from 200bp to 2kb. The alterations exhibited in the four conal cell lines included the loss of bands, shift of bands and intensity changes of bands. Conclusion Difference in genomic DNAs of four conal cell strains have been detected by RAPD analysis.

      Abstract:目的对用于实验动物皮肤病原真菌检测的分子生物学方法即皮肤病原真菌通用引物PCR与AP-PCR(任意引物聚合酶链式反应)相结合技术的特异性及敏感性进行评价,并通过建立动物模型及实际的皮肤真菌检测来验证此方法的可行性。方法特异性测定:用皮肤病原真菌通用引物进行PCR,扩增大肠杆菌DNA、犬肝炎病毒DNA及小鼠肝癌H22细胞DNA。敏感性测定:以紫外分光光度法测定DNA质量浓度,根据测定值将DNA进行10倍系列质量浓度稀释,得到100 ng~10 fg的8个质量浓度,然后对其进行PCR扩增。建立实验动物皮肤病原真菌感染的动物模型。用皮肤病原真菌通用引物PCR与AP-PCR相结合技术检测从动物模型及河北省各地的实验动物单位采集的动物皮肤标本。结果用皮肤病原真菌通用引物扩增实验动物三种皮肤病原真菌标准菌株,均扩增出大小为440 bp的条带,而大肠杆菌、犬肝炎病毒、小鼠肝癌H22细胞均未扩增出条带。皮肤病原真菌通用引物PCR敏感性测定,模板DNA 100 ng~100 fg的7个系列浓度均扩增出了阳性条带,10 fg为阴性。建立了实验动物皮肤病原真菌感染的动物模型。对河北省各地的实验动物皮肤真菌检测过程中出现一例通用引物阳性标本。结论皮肤病原真菌通用引物(CHS1 1S,CHS1 1R)具有较强的特异性,敏感度为100 fg;将皮肤病原真菌通用引物PCR与AP-PCR技术相结合可用于检测实验动物皮肤病原真菌。

      Abstract:目的探讨正常Beagle犬的生物学及心电图学特性参考值。方法正常Beagle犬200只,测量身长、体重、饮水量、进食量、心电图Ⅱ导联等指标。结果1.Beagle犬的体长、体重、进食量和饮水量有着相对稳定的对应关系。2.Beagle犬的心搏数雌雄略有差别,但P-R间期、QRS时段和P-T时限较为稳定。结论为正常Beagle犬的生物学及心电图学特性提供了参考依据。

      Abstract:Objective To establish an animal model which is similar to the pathological process of transplanted vein-bridge stenosis after coronary artery bypass graft(CABG).Methods Rabbit underwent interposition of the external jugular into the common carotid artery in a end-to-end fashion.Results One month after operation,lumen of vein-bridge stenosis was observed and histology exams showed that new intim was formatted.Conclusion This model can truly reflect the conditions of vein-bridge stenosis after CABG.

      Abstract:Objective To observe the fluctuation of uric acid in serum on mouse after replicating model by using hypoxanthine and uricase inhibitor. Make certain the most proper approach and solvent for using uricase inhibitor. Method Different approach and solvent were adopted for using uricase inhibitor, inspect the uric acid in serum at different periods of time. Results Hypoxanthine was administered via stomach, while uricase inhibitor was injected subcutaneously by lanolin mixed with olefin,which made the uric acid in serum higher than control group about 12 hours. Conclusion Uricase inhibitor was given subcutaneously by lanolin mixed with olefin in acute animal model with Hyperuricemia on rat, which has the characters of high uric acid and maintaining a long time.

      Abstract:In 2003, under the auspices of the main UK funders of biological and biomedical research, a working group was established with a remit to review potential welfare issues for genetically altered (GA) mice, to summarize current practice, and to recommend contemporary best practi.ce for welfare assessments. The working group has produced a report which makes practical recommendations for GA mouse welfare assessment and dissemination of welfare information between establishments using a 'mouse passport' . The report can be found at www. nc3rs, org. uk/GAmice and www. lal. org. uk/gaa and includes templates for the recommended welfare assessment scheme and the mouse passport. An overview is provided below.

      Abstract:目的研究SPF级近交系Wistar大鼠性别和年龄因素对其血常规值的影响。方法用MEK-7222K全自动血液分析仪检测SPF级近交系Wistar大鼠血常规值。结果SPF级近交系Wistar大鼠周龄对其体重、白细胞、淋巴细胞、单核细胞、血红蛋白、红细胞压积、血小板、血小板压积、血小板分布宽度有极显著影响(P〈0.01),周龄对中性粒细胞、血小板平均体积有显著影响(P〈0.05),周龄对嗜酸性细胞、嗜碱性细胞、红细胞、红细胞平均体积、平均红细胞血色素量无显著影响(P〉0.05)。SPF级Wistar大鼠性别对体重、白细胞、血小板分布宽度有极显著影响(P〈0.01),性别对嗜碱性细胞有显著影响(P〈0.05),性别对中性粒细胞、淋巴细胞、单核细胞、嗜酸性细胞、红细胞、血红蛋白、红细胞压积、红细胞平均体积、平均红细胞血色素量、血小板、血小板压积、血小板平均体积无显著影响(P〉0.05)。结论SPF级近交系Wistar大鼠的年龄、性别因素对其血常规值有显著的影响。

      Abstract:目的测定4~5周龄D2BF1小鼠血液学参考值,且与同龄DBA/2、C57BL小鼠比较,并提供D2BF1小鼠主要脏器重量、脏器系数的参考值。方法随机选取4~5周龄D2BF1小鼠雌雄各10只,分别称重,摘除眼球采血后,颈椎脱臼处死,摘除主要脏器称重,并将测定的血液学值与同龄的C57BL和DBA/2小鼠进行比较分析。结果D2BF1小鼠雌雄之间血液值MCV有显著性差异(P〈0.05),其余无显著性差异(P〉0.05)。与同龄的C57BL小鼠相比,血液值WBC、Lymph%、Gran%有极显著性差异(P〈0.01)。其余无显著性差异(P〉0.05)。与同龄的DBA/2小鼠相比,WBC、Mid%、RBC、MCH有极显著性差异(P〈0.01),Mid有显著性差异(P〈0.05)。其余无显著性差异(P〉0.05)。结论D2BF1代小鼠白细胞及分类明显低于C57BL和DBA/2小鼠,值得进一步研究。

      Abstract:目的通过评价芬太尼、司可林等药与戊巴比妥钠复合麻醉在急性心梗猪模型实验中的应用结果,建立一种较好的应用于大型动物实验的麻醉方案。方法28只猪被分成结扎组(AMIn=22)和假手术组(n=8)。AMI组动物开胸、结扎阻断心脏左冠状动脉的前降支(LAD 23、对角支处),假手术组开胸不结扎冠状动脉。手术动物使用氯胺酮(10 mg/kg)和阿托品(0.5 mg/只)肌肉注射进行诱导麻醉;开放静脉通道,术前缓注戊巴比妥钠(20 mg/kg)、芬太尼(0.008 mg/kg)进行基础麻醉;术中动物肌肉松弛采用司可林(5 mg/kg),并气管插管,联通呼吸机。结果使用该复合麻醉方案时,手术动物安静,手术中动物循环稳定,术后复苏迅速,手术实验质量提高。AMI组动物死亡率是20%;结扎后动物心电图S-T段明显提高;TTC染色:梗死区肉眼可见灰白色;光镜下:梗死区结缔组织纤维增生。结论该复合麻醉方案可以较好的应用于大型动物实验麻醉。

      Abstract:Objective To discusses the reason of generating short hairs character and try to inbreed the sparse strain. Method To design the genetic experiment by means of sib mating and back cross mating. Result The sparse mouse appeared in F2 of the inbreeding and later generations of the back cross mating with the race mouse x F~ . It was proved that the sparse character is related with heredity. Conclusion The sparse character is autosomal recessive inheritance which is controlled by monogenic, and its heredity rule is completely consistent with Mendelian inheritance.

      Abstract:Objective To establish a model of chronic atrophic gastritis in rat. Methods 20 mmol/L sodium deoxycholate and 30% ,60% alcohol were given in combinatin to rats to establish CAG model for 13 weeks. Pathologic changes of the gastric mucosa were observed by microscope, biochemical indexes were measured with instruments. Results Inflammatory index of gastric mucosa in CAG rats was significant increase, the thickness and area of inherent gland shrunk significantly ; the concentration of free acid and total acid were significant decrease, mucin contents and pepsase activity were significant higher, serum gastrin and malondialdedyeds contents enhanced significantly, interleukin-2 and superoxide dismutase activity decreased significantly. Conclusion Succeeded in establishment of an animal model of CAG, and provided an ideal animal model for the study of chronic atrophic gastritis.

      Abstract:Objective To study the function of ovary after ovary allotransplantation. Method The ovary allotransplantation of 10 days mice were performed to 1 month mice, which is the same strain or has the same genetic gene, the reproduction of the ovary receptors were observed, and PCR were used to test the little new-born. Result The result indicated that the transplanted ovary could recover, maintenance, and reproduce normal newborn, the newborns were come from the oocytes from the transplanted ovary. Conclution The ovary allotranplantation was successful.

    • Experimental Eye Irritation Caused by of 2% Peracetic Acid in Rabbits

      Abstract:Objective To determine whether 2% peracetic acid solution cause eye irritation in laboratory workers and animals. Methods 2% peracetic acid solution applied for a given time to the eyes in rabbits and the ocular changes were foUoweed and evaluated by score grading. Results Administration of 2% peracetic acid for 6 hours lead to iris and conjunetiva irritation, with hyleremia, edema and secretion. The changes were increased 24 h later, and serious changes were still present 7 days later. Conclusion 2% peracetic acid is a strong irritant and desinfection with 2% peracetie acid is irritative to laboratory animal eyes, suggesting that it is harmful to human eyes.

      Abstract:Objective To evaluate the feasibility of respiratory inhalation route on avian influenza virus(H5N1) in rhesus macaques.Method We inoculated rhesus macaques by thyrocricoid puncture and observed the clinical signs,pathologic changes and so on in a short period.Result Infection caused typical clinical signs and pathologic changes which were similar to human and animals infected with avian influenza virus.Conclusion Rhesus macaques could be infected by the inoculation of thyrocricoid puncture.And it was proved to be a good method for the similar experiments.

      Abstract:目的探索狗腰椎穿刺的方法,以便快捷顺利地安置导管监测颅内压、抽取脑脊液行生化分析等,为实验动物烧伤研究提供可靠的椎管内置管方法。方法20只重庆成年杂交犬行腰椎穿刺。术后对狗脊椎骨进行解剖学观察,测量后位腰椎棘突之间的夹角以及各棘突间孔的大小。并对烧伤早期脑水肿改变的实验犬行椎管内置放导管。结果对狗进行腰椎穿刺时应按以下步骤进行,先突破棘上韧带,用穿刺针弯头寻找下位棘突前缘,开始时穿刺针弯头朝向后,沿棘突前缘徐徐进针,达到棘突根部时突破黄韧带(椎弓与椎弓之间的韧带)有落空感,然后旋转穿刺针180°使弯头朝向前,再进针少许突破硬脊膜,拨出穿刺针蕊,伴有清亮液体缓缓流出。结论由于狗腰椎棘突与人有很大差异,本文展示了在狗腰椎安置导管监测颅内压的方法。

      Abstract:Rhesus Cytomegalovirus(RhCMV) is an endemic herpesvirus of rhesus macaques.It is closely related to human cmv(HCMV) in many characters,and many same clinical symptoms were observed in infected rhesus monkeys.So,it provides an excellent model for HCMV research.In this article,the authors review the RhCMV in terms of pathogenesis,immunity inhibit,embryo abnormality and detection methods.These contribute to develop new therapy measurement for the human diseases and provide a good evaluating platform for accelerating clinical progress,at the same time,it is very critical to species' protection.

      Abstract:Itching models have been used in studying the mechanism of itch and antipruritics. To establish these models, methods of applying special germ lines of laboratory animals, stimulated with mediators of inflammation or chemical materials and injection of morphine in central were used. The application of these methods in investigations of itch or antipruritics and the evaluation of itching model have been introduced.
