Abstract:目的研究阿尔茨海默病APP/PS双转基因小鼠自主行为的改变和作为阿尔茨海默病行为特征的可行性。方法对APPswe/PSΔE9双转基因小鼠和野生鼠分别在4月龄、6月龄、8月龄进行旷场自主行为实验,并与Morris水迷宫分析进行比较,利用SPSS16.0软件统计分析。结果在4月龄、6月龄转基因小鼠的学习记忆能力、以及自主性探究性行为与野生鼠比较有明显的差异。表现为:模型鼠学习记忆能力减弱,兴奋性增高,自主活动增加。这些表现与临床患者的症状有许多相似性,8月龄时,差异减小。结论APPswe/PSΔE9双转基因小鼠的自主行为学表现为烦躁不安,自主活动增加,与阿尔茨海默病患者的临床表现有许多相似之处,可作为小鼠模型判别的行为标志之一,在阿尔茨海默病的病因病机研究和药物研发等方面具有一定的应用价值。

Abstract:目的建立心脏特异表达Meox1转基因小鼠,研究Meox1对心脏发育及心肌病的调节作用。方法利用心脏特异启动子α-MHC构建转基因表达载体,显微注射法建立Meox1转基因小鼠,PCR鉴定转基因小鼠的基因型,Western blot检测Meox1在心脏组织中的表达,心脏超声检测转基因小鼠及野生小鼠的心脏结构和功能。结果在生理状态下,Meox1基因只在幼鼠心脏中表达,在病理状态下,Meox1基因在成年心肌病小鼠的心脏组织表达升高。通过显微注射,建立了两个Meox1基因在心脏组织的表达水平明显高于同龄对照小鼠的转基因小鼠品系。与野生型小鼠相比,两个Meox1转基因小鼠品系收缩期左室内径分别增加7.2%、12.8%(P〈0.01,n=16),舒张期左室内径分别增加15.6%、24.2%(P〈0.01,n=16),收缩期容积分别增加36.8%、65.7%(P〈0.01,n=16),舒张期容积分别增加18.2%、33.8%(P〈0.01,n=16)。射血分数分别减小6.6%、9.3%(P〈0.05,n=16),短轴内径缩短率分别减小9.4%、12.3%(P〈0.05,n=16)。结论Meox1在心肌病心脏中表达,其在心脏高表达引起心脏左室内径增加,收缩期容积和舒张期容积显著增大,射血分数及短轴缩短率减少等扩张性心肌病表型,是参与心肌病病理发生的基因之一。

Abstract:目的观察中等强度跑台运动对去卵巢大鼠骨质疏松的预防作用。方法将30只3月龄未经产雌性SD大鼠随机分为假手术、去卵巢静止和去卵巢运动三个组。去卵巢运动组每周进行4次时间45min、速度18m/min、坡度5°的跑台训练。实验结束时,检测血清雌二醇(E2)、碱性磷酸酶(ALP)、抗酒石酸酸性磷酸酶(TRAP)和骨钙素(BGP)水平以及右侧游离股骨和胫骨的骨密度(BMD)和骨矿物含量(BMC);同时观察左侧股骨远端和胫骨近端组织形态学变化。结果与假手术组比较,去卵巢静止组大鼠血清ALP活性和BGP含量显著升高,血清TRAP活性和E2含量显著下降,股骨近段和远端以及胫骨近端BMD和BMC显著下降,股骨远端和胫骨近端骨小梁断裂增加、数目减少;与去卵巢静止组比较,去卵巢运动组大鼠血清E2和BGP含量显著上升,股骨三个部位以及胫骨近端BMD和BMC显著增加,股骨远端和胫骨近端骨小梁断裂减少、数目增加。结论中等强度跑台运动能增加去卵巢大鼠血清E2和BGP含量,改善去卵巢大鼠骨组织学结构。

Abstract:目的探讨mir-106b在阿尔茨海默病发病中的作用。方法取6月龄APPswe/PSΔE9小鼠脑组织,进行microRNA芯片的检测;利用real-time PCR对芯片检测结果进行验证;构建mir-106b表达载体,将其转染至SH-SY5Y细胞中构建mir-106b稳定转染的稳转细胞系。用Targetsan、Pictar、miRanda等靶基因预测软件,对mir-106b的靶基因进行预测,根据靶基因的功能选择可能与AD发病相关的靶基因,并用Western blot对所选择的靶基因在稳转细胞中的表达进行验证。用双荧光素酶报告检测系统检测mir-106b与其靶基因的结合位点。结果芯片结果显示,与野生型小鼠相比,6月龄APPswe/PSΔE9小鼠脑组织中mir-106b的表达下降,经real-time PCR验证,mir-106b的表达在APPswe/PSΔE9小鼠脑组织中的表达较野生型小鼠升高,差别具有统计学意义(P=0.03)。mir-106b稳转细胞系较对照mir-106b的表达升高2~5倍。神经PAS结构域蛋白2(neuronal PAS domain protein2,NPAS2)在mir-106b稳转细胞中的表达明显下降。双荧光素酶报告实验证实:与对照相比,带有NPAS2的3'UTR的荧光素酶报告载体与mir-106b表达载体共转染,海肾荧光素酶/萤火虫荧光素酶的相对活性降低;荧光素酶报告载体的NPAS2-3'UTR突变后与mir-106b表达载体共转染,海肾荧光素酶/萤火虫荧光素酶的相对活性升高。结论mir-106b可能通过调节机体生物钟基因NPAS2的表达,影响阿尔茨海默病人的生活节律,参与阿尔茨海默病的发生。

Abstract:目的S100B在多种神经损伤性疾病中高表达,高浓度的S100B蛋白对中枢神经系统具有毒性作用。建立脑组织特异表达S100B转基因小鼠,用于研究该基因在帕金森病(Parkinson's disease,PD)发病中的作用。方法ELISA法检测S100B蛋白在α-synuclein A53T转基因小鼠脑组织中的表达情况。构建PDGF-hS100B表达载体,显微注射法建立S100B转基因小鼠。PCR鉴定转基因鼠的基因型,Western blot检测基因表达水平。Rota-rod实验检测S100B转基因鼠的运动协调能力。结果S100B在9月龄和12月龄α-synucleinA53T转基因小鼠脑组织中的表达高于野生型小鼠。建立了2个品系的脑组织特异表达S100B转基因小鼠。与野生型小鼠相比S100B转基因小鼠表现出明显的进行性运动协调能力障碍。结论S100B转基因小鼠的建立为研究该基因在PD中的作用提供了工具。

Abstract:Pet animals can habor and transmit many zoonotic pathogens.Many factors drive infections,such as frequent household pets increasing in numbers and species,and the booming of international trade of exotic pets.This review summarized the zoonotic pathogens potentially transmitted by pet dogs,cats,rodents,birds,fish and reptiles to humans,and described the route of infection and the zoonoses caused by these pathogens.The preventive strategies for petassociated infections were summarized.
