    • Analysis of Heart Function of the Heart-specific Expression Calponin 1 Transgenic Mice

      Abstract:Objective To establish the heart-specific expression Calponin 1 transgenic mice and investigate its effects on the development of heart and cardiomyopathy. Method The transgenic vector was constructed by inserting the human Calponin 1 gene into the down-stream of α-MHC promoter. The transgenic mice were generated by the method of microinjection. The genotype of transgenic lines was identified by PCR,and the expression levels of the Calponin 1 gene were detected by Western Blot. The pathologic changes of the heart structure and function were analyzed by echocardiography,hematoxylin and eosin( HE) stain and Masson stain. Result The expression of Calponin 1 was detected in the heart of different age wide-type mice by Western Blot,but was down-regulated in the mice with DCM. Compared with the wild type mice,transgenic mice showed significant heart remodeling,left ventricular systolic diameter increased by28% (P < 0. 01,n = 12) ,left ventricular diastolic diameter elevated by 16. 2% (P < 0. 01,n = 12) ,left ventricular posterior wall during systolic decreased by 15. 7% (P < 0. 01,n = 12) ,and left ventricular posterior wall during diastole reduced by 21% (P < 0. 01,n = 12) . Transgenic mice,versed wild type,showed weakened heart function,ejection fraction diminished by 11. 5% (P < 0. 01,n = 12) and fraction shortening reduced by 14. 6% (P < 0. 05,n = 12) .The heart of the transgenic mice exhibited an enlarged ventricular chamber when compared with that of the wild type with HE stain and Masson stain. Conclusion Over-expression of Calponin 1 in the heart of the transgenic mice caused a phenotype similar with dilated cardiomyopathy. It suggested that Calponin 1 might be one of the modifier genes which involved in the pathogenesis of cardiomyopathy.

    • The Infection of Fmr1 on Physiological Development and Reproductive Performance in Mice

      Abstract:Objective To investigate the indices of physiological development and reproductive performance in FVB. KO and FVB. Method 40 seeds FVB. KO and FVB (20 male and 20 female mice,aged 10 weeks) were chose and monogamously mated. After natural delivery,the research tested the reproductive performance and indices of physiological development such as pinna detachment,body hair sprouted,incisor eruption eye opening. Result The reproductive performance and indices of physiological development but rate of male and female in FVB. KO and FVB were no difference.The rate of male and female in FVB. KO was lower. Conclusion Fmr1 has no effect on behavioral development and reproductive performance in mice. But it maybe has some effects on rate of male and female in mice.

    • The Influence on Wistar Rat’s Organs with Repeated Blood Collection

      Abstract:Objective To study how the chief organs of male Wistar rats were affected after a number of blood collections through their orbital cavity at different intervals. Method The experimental group had blood collection with an interval of 3 days,7 days and 10 days respectively. The blood was collected through orbital venous plexus and with once 1 mL,3 times totally. The control group had once blood collection after the experimental group got it the last time with only 1 mL. Their weights were measured when blood was collected and the Wistar rats were killed after all the collections had been done with their organs ( heart,liver,kidney,spleen,thymus) being weighted the coefficient of the organs. Result The blood collection was done with an interval of 3 days. The significance existed in the rat’s liver and kidneys’coefficient( p< 0. 05) . If the blood was collected with an interval of 7 days and 10 days,there was no any significant difference.Conclusion The blood collection with an interval of 3 days has an influence on the rat’s liver and kidney coefficient and that of 7 days or 10 days has no influence on them.

      Abstract:Ebola virus,which was first identified in 1976 during outbreaks in Ebola River vally,is an zoonotic pathogen that causes highly lethal hemorrhagic fever syndrome in human and nonhuman primates. Since the high mortality rates of Ebola viruses,up to 88% in humans,Ebola viruses are listed in the most dangerous viruses to humans by World Health Organization. Comprehending the characterization and pathogenesis of Ebola hemorrhagic fever and Ebola viruses,is very important for the prevention and control of this disease.

      Abstract:Hydrodynamics-based transfection is a kind of exogenous gene transfection. It’s a fast,convenient and high-level expression way. We injected a great deal of normal saline concluding the target gene into mice through caudal veins quickly,the exogenous gene could express highly in vivo. This way has already applied in animal models and gene therapy of liver-disease,myocarditis,nephropathy and anti-tumor. In addition,this way had widespread used in gene function,gene regulation,protein expression and gene therapy in vivo.

    • Influence of Different Music Environment on Growth and Development of Mice

      Abstract:Objective To investigate the influence of music on growth and development in mice. Method 30 ICR mice which were fed 27 days in music environment of 50 dB and 60 dB of Guzheng and Hulu hereby,were weighed once per 3 days,and were measured digestibility of various nutrients of feed once per 9 days. Result The body weights of female mice in different music environment were all higher than control group (P < 0. 05) ,and 50 dB groups were higherthan 60 dB groups in all music environment (P < 0. 05) . The body weights of male mice in Guzheng music environment were higher than that of control group ( P < 0. 05 ) . However the body weights of male mice in Hulu hereby music environment were lower than control group. The digestibility of various nutrients of mice had no significant difference in different music environment (P > 0. 05) . Conclusion Different music environment have a significant effect on the body weight of mice,and have no effect on the digestibility of various nutrients.
