Abstract:Both ‘Human Genome Project ( HGP ) ’ and animal cloning from somatic cells will be listed as two important milestones in the history of life sciences. The success of HGP started the new era of genomics with two major perceptions that“life is in sequence”and“life is digital”. Based on this belief,BGI has been focusing on sequencing and bioinformatics,aiming at analyzing genomes of “everybody in the world”and“every( living) thing on Earth”via vast collaborations. Since the birth of Dolly the sheep,successes have been made in cloning other mammals. With the main principle of reprogramming,somatic cell nuclear transfer ( SCNT) is becoming the bond among genetic munipulation,stem cell and synthetic biology,making the 20th Century “The Century of Biology”. Based on the information obtained from genomics and improved technology of handmade cloning,BGI has established an animal cloning platform,which focuses on: 1 ) generating animal models for human diseases as a strong tool for studying the mechanism and developing therapy methods of human diseases; 2) livestock improvement for making life quality better.

Abstract:Experimental animals are an important tool and supporting condition for research,teaching,production,standardization. The species and strain of experimental animals used in research have been increased markedly alone with development at full speed of science and technique. But the biological diversity,ecological environment and human health have been threatened by experimental animals bio-safety issues in our country. This review analyzed the potential bio-safety hidden danger in experimental animals raising and use,investigated problems existed in experimentalanimals bio-safety control,and proposed some measures and advice to laboratory animals bio-safety.

Abstract:Many species and a large number of experimental animals have been widely used with a long history in the control tests and development of veterinary biologicals. The existing problems related to the animals used in the control tests or research of veterinary biologicals are discussed in the article and some suggestions on the establishment of large experimental animal bases to guarantee supply,improvement of the quality standards to guarantee quality,renewal of thought and improvement of laws and regulations to decrease the number of experimental animals used and reinforced management to ensure biosafety and biosecurity have been presented.

Abstract:Laboratory animal science is an essential element to support life science research. Its industrialization has been recognized as an important indicating and plays an important role in the development of science technology and economy in a country. The industrialization of laboratory animal science consists of laboratory animal breeding and supply,related products of laboratory animals,service of laboratory animal experiments,which serves for many fields of scientific research. The laboratory animal industrialization was initiated only for few decades. With the development of life science research and the improvement of comprehensive national power in China,the laboratory animal industrialization was development rapidly in the country. However,several problems remain to be resolved in the field in China: 1) China lacks of companies with big scale and great influence in our country even in the world; 2 ) The quality of products/service remains to be improved; 3) The system of quality management and supervision remains to be strengthened; 4) China lacks of professionals of the field. According to the problems,the following strategies are suggested so as to improve the development of laboratory animal sciences as well as human being health in China: 1 ) To provide powerful policy and financial support by government,so as to found several large-scale companies in the field; 2 ) To strengthen the development of quality management and supervision system,so as to improve the quality of products and services; 3) To pay more attention to fostering human resources,so as to improve the capability of the supply and services.

Abstract:Laboratory animal medicine ( LAM) ,as a specialty within veterinary medicine,started in 1950s in the USA. USA represents probably the most advanced LAM in the world. The objective of this article was to concisely brief the history of LAM in the USA,including the needs for laboratory animal veterinarians in Biomedical field; early veterinary practice in laboratory animal sciences; the establishment of American College of Laboratory Animal Medicine ( ACLAM) ;the historical and current practice of education,training and certification in LAM; and the impact and influence of laws and regulations on LAM. Based upon personal experiences of the authors and the national situations in China,it was proposed that strategy for LAM development in China would include but not limited to the followings; further consummating laboratory animal related laws and regulations,strengthening LAM contents in veterinary education and training,advocating an promoting the training of future LAM leaders,enhancing multi-channel networking among LAM professionals,further supporting and investing in LAM related research. “Standing on the shoulders”of well developed LAM collective wisdom,we should be able to build up,step by step,our LAM specialty with Chinese characteristics.

Abstract:In recent years,the health status of laboratory rodent colonies is dramatically improved. However,there are still quite a few important infectious microorganisms staying prevalent or emerging in the laboratory rodent colonies. The health of laboratory rodents affects their suitability for research,accuracy and integrity of research data,animal welfare and human health. Therefore,an animal health quality control program,often termed health surveillance,health monitoring,is important to any research facility. Based upon authors’hands-on experiences,the basic principles of health monitoring program were summarized as follows; the necessity of health monitoring,how to develop an effective health monitoring program,sentinel program,common testing methodologies,sampling strategy,and how to interpret and respond to health monitoring results. An effective and practical health monitoring program should be established based on those aforementioned principles,and being consummated during implementation.

Abstract:Research biologics are widely used in biomedical research. Contaminations of research biologics with infectious agents are risk factors for vivarium biosecurity and may compromise the in vivo research. Therefore,precautionary screening on the biologics should be perform,before inoculating those biologic materials into rodents,in order to confirm that they are free of rodent infectious agents. Due to scientific,technical and economic constraints,it is not feasible to screen for all rodent infectious organisms. This overview was a proactive attempt,based upon the current literature,personal experience and expert opinion,in trying to describe how to set up a strategy for microbiologic contamination screening and the rationale for selecting certain infectious agent for testing. The impact of those agents on research,vivarium biosecurity,and occupational health and safety,is also summarized.

Abstract:Results of quality control test and even the quality of the products are directly affected by SPF chickens and SPF embryos ,which are the main raw materials of production and quality inspection in veterinary biological products.Chickens and embryos must meet the national SPF animal standards requirement from the animal standards in production and inspection in the veterinary pharmacopoeia of the People's Republic of China ( 2010 ) ,in the seed lots and biological products preparation and quality inspection. The quality control of SPF chicken includes four tests,namely inheritance,microorganism ,environment and nutrition respectively. The current standards for quality control of SPF chickens are reviewed in the perspective of bioproducts enterprises. In this article,critical ideas and practical proposals for quality control of SPF chickens based on the practice of production and quality inspect in poultry live vaccine are presented.

Abstract:Experimental animals and animal experimentation played a crucial role in the development of biological and modern medical research. The rapid development of laboratory animal science in China was associated with a growing demand for veterinary services to meet the demanding requirement for better medical care to laboratory animals. In this paper,we discuss the role of veterinary in animal experimentation. This article discusses the required qualifications for laboratory animal veterinarians,their fundamental missions and role in the health management of laboratory animals,and emphasizes the importance of the responsibilities veterinarians are entrusted with for the care of laboratory animals.Laboratory animal veterinarians play a key role in the health management of laboratory animals and the conduct of animal experimentation. Abstract:According to the World Health Organization,major depression disorder is projected to reach second place as leading contributor to the global burden of disease by the year 2020. Despite the emergency need for better therapies of depression,and major advances in the understanding of the molecular basis of this disorder in recent years,efforts to discover and develop new drugs for major depression disorder ,especially those which might revolutionize disease treatment,have been relatively unsuccessful. An important reason for this is that it is exceedingly difficult to build an animal model that perfectly recapitulates the symptoms of depression in human patients. In order to enhance clinical validity of animal models of depression,we reviewed the recent animal models of depression and discussed its translational research here.

Abstract:bjective To observe the regulatory effect o f BIOSTIME probiotics on intestinal flora. Method Mice were divided into normal control group,BIOSTIME probiotics group. Control group fed with distilled water for 14 d.The BIOSTIME probiotics group fed with BIOSTIME probiotics for 14 d. The numbers of intestinal flora before and after experiments were detected. Result The numbers of intestinal Bifidobacterium of the groups fed with BIOSTIME probiotics superior to those of normal control group( P < 0. 05) ,while the number of E. coli,Enterococcus ,Lactobacillus,Clostridium perfringens was not significantly changed( P > 0. 05) ,compared with the data prior of feeding. Conclusion BIOSTIME has a regulatory effect on mouse intestinal flora.

Abstract:Objective Chemotherapy-induced peripheral neuropathy ( CIPN) is the common adverse reactions of mutiple chemotherapy drugs. At present,CIPN model is most commonly established by injecting paclitaxel ( PTX ) intraperitoneally to SD rats. However,those methods differ a lot and the influence factors are still unknown. Thus in this paper we aim to find the influence factors and optimize an ideal model for CIPN research. Methods SD rats were injected i. p. every other day with PTX. Mechanical allodynia and mechanical hyperalgesia effects were examined by the behavioral test. Intraepidermal nerve fiber ( IENF) of the skin of hind foot was stained by immunofluorescence and morphology and number of mitochondrial on the sciatic nerve was detected with electron microscopy. Results Injection of 2 mg·kg - 1 PTX for 4 times had no effect on the weight of SD rats. No obvious mechanical abnormalities pain and mechanical allodynia was observed during the 34 d experimental duration in rats with the initial weight of 200 g. While in rats with the initial weight of 400 g,mechanical abnormalities pain was observed at the 17d,and mechanical hyperalgesia lasted from the 8 d to the 26 d. At the same time,density of IENF under the skin was significantly reduced after PTX treatment,and the proportion of abnormal mitochondria was also increased,showing severe peripheral neuropathy. Conclusions Rats with initial body weight of 400 g are more sensitive to establish PTX-induced peripheral neuropathy and are more suitable for CIPN research.

Abstract:Objective Collagen induced arthritis ( CIA) is an ideal model for studying the mechanism of rheumatoid arthritis ( RA) and exploring the anti-RA drug candidates. However,there are few studies about the systemic immune abnormality of CIA mice as yet. Thus,we established CIA model using collagen II and evaluated it. Methods Blending collagen II and Complete Freund's adjuvant and emulsifying them completely,then injected them to male DBA /1 mice at root of the tail. After 20 days,repeated this process. The swelling of the claw were determined using micrometer.ELISA,Luminex and αLISA were employed to measure the collagen II-specific antibodies and multiple cytokines. Results Beginning from 23 days of injection,red swelling of the claw skin appeared in some mice. At the end of experiment,100% mice developed arthritis symptoms. The spleen index of CIA mice increased significantly,accompanied by augmented spleen lymphocyte proliferation reaction to collagen II. At the same time,multiple cytokine levels in the supernatant of cultured spleen lymphocytes increased than control,such as IFN-γ,TNF-α and IL-1β. In addition,there was high amount collagen II-specific antibodie in the serum of CIA mice. Conclusions CIA mice characterized by abnormal activation of T cell,especially Th1 cell,with a high incidence,is a suitable RA model

Abstract:Depression is a psychological disorder characterized by behavioral, neurochemical and other physiological abnormities. Chronic exposure to mild unpredictable stress( CUMS) has previously been found to cause an antidepressant-reversible decrease in the consumption of palatable sweet solutions, at the same time biochemical disturbances induced by the CUMS procedure were similar to those found in human depression. We review the pathological changes of the animal model of depression ( CUMS ) . it reflected in some persistent molecular pathologys,such as the changes in receptor ,signal transduction.

Abstract:Objective To realize the potentially developmental neurotoxicity of chlorpromazine ( CPZ ) on zebrafish embryos and Larvae. Methods The effect of the general toxicity in zebrafish embryos and larvae were assessed after high doses of CPZ ( ≥5 mg /L) administered to zebrafish embryos on 6 - 72 hours post fertilization ( hpf) . Then,pathology change and cell apoptosis were investigated. Under teratogenic dosage of CPZ ( ≤2. 0 mg /L) administered to zebrafish embryos on 6 - 72 hpf,the escape times of 3 dpf larvae in the touched -evoke escape reaction was recorded.Moreover,spontaneous movement and light-evoked startle escape response of 6 dpf zebrafish were tracked and analyzed by using a video-tracking system. Results The lethal and teratogenic effects of CPZ was dose-dependent and stagedependent. Histopathological examination revealed that brain volume and brain cells of 7 dpf larvae have reduced in size.The number of apoptotic cells of 36 hpf embryos has increased in midbrain,hindbrain,hypothalamus,midbrain hindbrain boundary,notochord as well as tail. The weaker tactile sensitivity was displayed when 3 dpf larvae were exposured to CPZ ( ≥0. 125 mg /L) . Hypoactivity was discovered when 6 dpf larvae were exposured to CPZ ( ≥0. 5 mg /L) . Furthermore,CPZ also evokes tremor,freezing or stereotypic circling swimming in 6 dpf larvae. CPZ ( 1. 0 mg /L or 2. 0 mg /L) can inhibit but CPZ ( 0. 125 mg /L or 0. 25 mg /L) can promote hyperactivity by light-evoked startle reaction. Conclusion CPZ can cause developmental neurotoxicity on zebrafish embryos and larvae. In addition,zebrafish larvae have obvious advantages to make it a powerful alternative for the developmental neurotoxicity assessment of exogenous compounds by using high-throughput behavioral test methods.

Abstract:Objective To obtain the basic data of the Sprague-Dawley rats in embryo-fetal development toxicity study. Methods Timed-pregnant rats ( n = 395) were sacrificed at the end of the gestation period ( GD 20) . Terminal examinations include necropsy,count of corporea lutea and live and dead implantations,gross evaluation of placenta,fetal body weight,and external,visceral,and skeletal examination of fetuses. 50% of fetuses were examined for visceral alterations and 50% of fetuses for skeletal abnormalities. Results and Conclusion The basic data of the Sprague-Dawley rats,such as body weight,fetal development,and fetal malformation / variation,were obtained.

Abstract:In this paper we present the conceptions: Laboratory animal welfare is that human should give good treatments to laboratory animals ( positive factors) ,which is mainly performed in animal breeding and transportation with “5F”as the basic theory,while ethics is of human should follow the principles in animal experiments ( negative factors) ,which is mainly carried out in the animal experiment with“3R”as basic theory. We also summarized the specific practices to keep welfare of laboratory animals breeding and transportation,such as environment,cages,bedding,density,feed,water,social and behavioral need,euthanasia,transportation,etc.

Abstract:Objective To improve the consciousness of biosafety in livestock laboratory,and put forward the experience for reference of design,construction,application and management of livestock biosafety laboratory. Method It is investgated and analyzed that designing idea, equipment, facilities, animal experiments and livestock laboratory management in North American. Results The livestock biosafety laboratory is related to healthy progress of animal husbandry and social stabilization. The construction of livestock biosafety laboratory between home and aboard exists remarkably different. Conclusion We should use the experience of designed idea in building livestock biosafety laboratory of North American for reference,consummate the infrastructure of livestock biosafety laboratory and add corresponding equipment,and improve the biosafety consciousness of animal laboratory staff simultaneously.
