Abstract:ObjectiveTo explore the relationship between gene expression of Cricetulus barabensis TYR gene family and Cricetulus barabensis coat color. Methods The relative expression quantity of TYR, TYRP1 in Cricetulus barabensis of different colors was analyzed by using real time quantitative PCR in this research. ResultsThe relative expression quantity of TYR, TYRP1 in Cricetulus barabensis respectively were 2.5,5.3 times than that in the Albino mutant, all results were corrected by the household gene. Conclusion The findings indicated that gene expression level of TYR gene family in Cricetulus barabensis were higher than that in the Albino mutant, and the gene expression level of TYR gene family were related with phenotype of Cricetulus barabensis coat color.

Abstract:ObjectiveTo detect the function of I BHV-1 TK-/gE-/ EGFP+ gene-deleted mutant virus strain, find a new method to prevent infectious bovine rhinotracheitis (IBR), the charaction of TK-/gE-/ EGFP+ gene-deleted mutant virus strain was further studied. Methods Southern hybridization, western dot blot and Plaques test were employed in this investment. ResultsTK gene is deleted and the mutant cannot produce glycoprotein E when gE gene is deleted. The diameter of mutant strain much smaller than that of field strain, the mutant strain grows on MDBK cells and the virus titer can keep 106.0TCID50/0.1 mL for more than 40 hours, which indicated that the mutant strain inherit instantly. Conclusion The gene-deleted mutant virus strain was successful constructed in this study, and the mutant virus strain was stable. This will be useful for the further study of IBR.

Abstract:Objective To study the antihypertensive effects of s corn protein and its hydrolysates on spontaneously hypertensive rats (SHR).MethodsSixty 6~8 week-old male SHR were randomly divided into 5 groups:group A (corn protein), group B (corn protein''s hydrolysate prepared with pepsin), group C(corn protein''s hydrolysate prepared with trypsin), group D (corn protein''s hydrolysate prepared with complex enzyme) and group E(saline). Each group were orally adiministered at dose of 1g/kg body weight. Subsequently, systolic blood pressure of SHR was measured at a fixed time interval (0,1, 2,4, 6 and 8 h) post-adiministration. ResultsResults showed that all hydrolysates had significant inhibition on blood pressure of SHR and compound enzyme hydrolysate had the strongest effect on antihypertension during 0~6 h, with SBP decreased significantly which compared with group A, B, C and E (P<0.01). ConclusionA significant decrease in systolic blood pressure of SHR was observed in group, B, C and D.

Abstract:ObjectiveTo construct an engineering strain of pMAL- C2x - Tα1/TB1 which can express Thymosin α1(Tα1).MethodsTα1 gene was synthesized and amplified by PCR, PCR product and pMAL- C2x vector were digested with restriction endonuclease BamHI and EcoR I respectively, and were linked with T4 DNA ligase to construct pMAL- C2x - Tα1.After being sequenced, pMAL- C2x - Tα1 vector was transformed into E.coli TB1 for fusion expression under induction of IPTG.. The expressed product was identified by Western blot. ResultsThe DNA sequence of the synthesized Tα1 gene was identical to the original design.The constructed recombinant plasmid pMAL- C2x - Tα1 was highly expressed in E.coli TB1. The BMP - Tα1 fusion protein was about 33.6% of the total bacteria protein and the relative molecular weight of BMP - Tα1 fusion protein was about 45×103.ConclusionThe successful construction and expression of pMAL- C2x - Tα1/TB1 laid a foundation for the research of purification and biological acitivity of recombinant Tα1.

Abstract:ObjectiveTo compare Comparative study of propofol intravenous anesthesia and isoflurane inhalation anesthesia in Bama Minipigs.MethodsBama Minipigs 10, the average into two groups are propofol intravenous anesthesia and isoflurane inhalation anesthesia, and in preoperative, preoperative and postoperative the anesthesia monitoring. ResultsTwo groups of weight number of pigs, operation time and anesthesia time no significant difference of difference (P>0.05); Different fluorine ether group restore spontaneous breathing a short time in propofol group (P<0.05); Compared with the basic value, the experiment in anesthesia pig increased significantly after all HR value (P>0.01), reduce the MAP is obvious (P>0.01); But each rooms and in the group SPO2 and PH value difference was not significant (P>0.05).ConclusionPropofol intravenous anesthesia during the operation should be based on the experimental animals of the reaction conditions appropriate adjustments propofol pump into the concentration; and different fluorine ether inhaled anesthesia during the procedure of smooth, anaeesthetic effect is good, postoperative came quickly, suitable for complicated cases, and the time is long operation.

Abstract:ObjectiveTo compare the difference between the electroretinogram (ERG) normal rhesus monkeys and humans, and provide a base for further study on the application of rhesus monkey retinal disease as an experiment mode1. Methods The electroretnogram (ERG) was recorded in 7 normal rhesus monkey (14 eyes) and 8 human (16 eyes). The recording items of ERG included rod response b wave, cone response, max response a and b waves, oscillatory potential O2,0Hz flicker ERG ΣP. The results were statistically tested. ResultsThe ERG waveform results in rhesus monkey are similar to those in humans, but the amplitudes were decreased and the implicit times were prolonged compared with the humans, and the differences in the amplitude of the Cone ERG and the flicker ERG were not statistically significant. ConclusionsThe retinal function in rhesus monkey is quite similar to those in humans in the ERG recording. Rhesus monkey is a good animal model for the study on human retinal disease especially the macular.

Abstract:ObjectiveTo clone and identify alternatively spliced variants of beagles estrogen receptor β.MethodsBy RT-PCR analysis, we identified the existence of a new splice variant of beagles estrogen receptor β using Beagles pituitary organization as a template, to determine the existence of a new spliced variant by electrophoresis and cloned and sequenced. ResultsUsing primers designed to get two obvious electrophoresis with sequencing showed that one of the missing splicing, this alternatively spliced variants has not been reported.ConclusionWe get a new estrogen receptor β, this will help to study protein-coding function of a variety of estrogen receptor variable spliced and the role in the process of reproductive regulation.

Abstract:ObjectiveTo isolated and identify a bacterium from Bama minipigs of diarrhea and the patent medicine were screened by drug susceptibility test.MethodsA bacterium isolated from Bama minipigs was identified by morphological, germiculture, staining, Molecular biology and biochemical characteristics. The resistance test were used to selected the sensitive medicine by 25 drugs. ResultsThe Colibacillus of Bama Minipigs were success isolated and identified as serotype O138. The result of resistance test to 25 drugs showed that the bacterium was sensitive to Penicillin、Streptomycin、Rocephin and Nitrofurantoin, but highly resistant to another drugs that were used.ConclusionThe animal regression test showed that the bacterium have high pathogenicity. The result of this paper would provide the theory basis for prevention and control Colibacillus Infections of related experimental animal.

Abstract:ObjectiveTo compare the uterine and ovarian morphologies between diestrus and estrus Beagles for the further study of their reproductive inefficiency.Methods Detected the serum sexual hormone of 23 Beagle bitches, which have been produced once or twice, using chemiluminescence and selected 2 estrus and 4 diestrus Beagles by serological identification for ovarian and uterine specimens, 4% neutral paraformaldehyde-fixed, paraffin-embedded, routine HE stained, photographed. ResultsThe uterus and endometrium of diestrous dogs are thin, interstitial fibrosis, corpus luteum are occupied by primary follicles, 1-2 visible secondary follicles, no mature follicles. Follicular and luteal cells have more fiber, less vascularized. While the uterus and endometrium of estrus Canines are more thicken, endometrial cavity is larger, part of the gland is branched bend, glandular cells are enlarged, cytoplasm pale dyed, with a small number of visible vacuoles under the nuclear. Ovarian follicles are in large numbers, and are mainly primary, secondary and 1-2 mature follicles. There are numeric luteal cells which arranged regulatory, state clearly, loose interstitial fibrosis, vascular, and no vacuolar degeneration. Diestrus and estrus dogs’ ovarian have no obvious white body. Conclusion Diestrous Beagle sex hormone is maintained at a relatively low level, in proestrus estrogen levels increased rapidly, and after ovulation progesterone levels were higher. Morphological changes of ovary and uterus are adapted to the estrous cycle of bitches.

Abstract:Objective To explore the folic acid and adult neural stem cells’ joint influencing mechanism on the rat with Traumatic Brain Injury, and to find out its possible mechanism. Methods Divide 120 rats to 6 groups randomly—normal group, mode group, sham operation group, folacin injection group, adult stem cells transplant group, and folacin injection plus adult stem cells transplant group. Observe the morphological change under the microscope, then, do flow cytometry test and detect the expression of the neural stem cells’ facial notation—CD105、CD45、CD44、CD29. Examine the expression of neuron special enolase( NSE) and the expression of gelatinous fibre acidic protein (GFAP) with immunofluorescence. Examine the rats’ ability to motor-coordinate and conform with balance beam. Test each group’s learning and memorizing ability by conducting the Morris water maze experiment. Then, conduct the HE chromosome and Brdu immunohistochemistry experiment to detect morphological change of the brain tissue. After this, do the enzyme-linked immunosorbent assay (ELISA) to detect the expression of brain-derived neurotrophic factor( BDNF) and the expression of nerve growth factor. The last but not least, use western blotting to examine the expression of related dead protein in the brain tissue:BCL-2、Bax、Caspase-3. ResultsThe separated cells can be vitro subcultured, by doing flow cytometry test, we find that positive cells express CD44 and CD29 while negative express CD105, CD45. Cells induced by fetal bovine serum can produce NSE or GFAP positive cells. Experiments suggest that traumatic brain injured rats can significantly improve their behavior after jointly influenced by the folic acid and adult neural stem cells. Besides, they can also reduce brain tissue inflammation, restore damaged nerve cells, increase brain tissue of BDNF and NGF''s content, increase BCL 2 expression, and lower the expression of Bax, caspase-3. Conclusion The folic acid combined with adult neural stem cell intervention in traumatic brain injury in rats can significantly improve the central nervous system dysfunction, plays an important role to maintain the steady microenvironment in the neuron.

Abstract:Objective To investigate the morbility of MODS in different altitude and observe the change of blood biochemistry by the animal model of multiple orgen dysfunction syndrome (MODS) induced by lipopolysaccharide (LPS) in sheep.Methods 12 sheep were randomly separated into 2 groups, 6 sheep each group:the highland region of Sangke prairie (average altitude 3100 m) , sub highland region of Lanzhou group (average altitude 1520 m). After anaesthetizing , the MODS model was established by infusing 6 μg / kg LPS in 30 minutes intravenously in sheep. Then the physical sign appearance was observed and the indexes of blood biochemistry were determined in each group at different times.ResultsThe sheep in Sangke group, the death rate was markedly raised. The CK、CK-MB、AST、ALT、CRE biochemical indexes was markedly higher compared with Lanzhou group, except ALP that was little change. Conclusion MODS has the more damage in multiple organ follow on the raising altitude at highland.

Abstract:This paper mainly expounds a large complex low pressure tank in the development and the application in medical experiments. This tank can simulate a single climate conditions, and also can simulate various site of the composite climate environment through regulation. Abstract:Diabetic Nephropathy is well known as complication of microvessel disease caused by diabetes. High glucose leads to compensable high filtration rate of kidney,and then contribute to kidney cell groth, inflammation and fibrosis. Farnesoid X Receptor has been proved to ameliorate diabetic nephropathy. FXR can improve diabetic nephropathy through different aspects such as glucose, lipid, inflammation and fibrosis. How FXR improve diabetic nephropathy will be discussed in this review.
