    • Simple Analysis of Euramerican Laboratory Animal Welfare Legislation

      Abstract:The laboratory animals sacrifice for scientific-technical progress and human health. In the recent decades, ethical issues which associate with animal's quality of life have increasingly become a topic of public policy and controversy. Laboratory animal welfare laws can regulate human action, and give laboratory animals the legal rights and status as they deserves. In Euramerican developed countries, laboratory animal welfare laws and regulations are relatively perfect, especially in the UK, it has the most animal welfare legislations in the world, and has a great influence to the world. In-depth study of Europe and America advanced law is of great significance to the legislation of our country.

      Abstract:Allergic dermatitis is the skin diseases caused by allergen, is mainly refers to skin irritation, itching and peeling after exposure to certain allergens. At present, the sensitizing potential of chemicals is usually identified and characterized using animal test methods. Due to the increasing public and political concerns regarding the use of animals for the screening of new chemicals, using in vitro cell lines instead of animal models has become a trend. The human cell line activation test is mainly by detecting changes in cell surface markers and in the signal molecules after exposure to chemicals. The issue has been verified its feasibility by many laboratories in Europe, America and Japan. But at present the clinical value has not yet been approved. After some further optimization, new markers and methods are being developed and validated.

      Abstract:Objective To understand the biological characteristics of congenital cataract rats. Methods Expanding populations by breeding with the original group, then to observe reproductive performance and offspring growth and development. Results There was no significant differenceboth in the birth rate and the weaning number between the normal controlled group and cataract and microphthalmos cataract groups (p>0.05). Compared with other two groups,the cataract group had heavier weight at the second week after birth.In comparison between males of different groups, the cataract group growed faster at first two weeks after birth and the difference in weight was more obvious at fourth week after bieth(p<0.01) .In comparison between sexes, there was significant differencesin in weight between male and famale in the cataract group and the control group at the fifth week after birth (p<0.01), while the microphthalmos cataract group had significant differences at the sixth week (p<0.01). Conclusion Cataract gene in rats did not affect their reproductive rate, while the growth rate of pups had a certain impact.

    • The Effects of High Fat Diet on Joints of Collagen-induced Arthritis Rats

      Abstract:Objective To study the effects of high fat diet on joints of collagen-induced arthritis rats by comparing different diets. Methods After the model of Wistar CIA rats was established, We feed normal and high fat diet. The arthritis index scores were measured and the synovium were examined with HE stain and the concentrations of lipids and inflammatory factors in serum were detected. Results Compared with those in normal group, the arthritis index scores, synovial proliferation, pannus formation and inflammatory cell infiltration in synovium HE stain were increased in CIA rats with high fat diet. Plasma levels of TC and LDL were also enhanced in high fat diet CIA rats group(P<0.05) apart from HDL, which were decreased obviously(P<0.05).Plasma levels of Pro-inflammatory cytokines,that is TNF-α, IL-1β and IL-17 were also significantly increased in high fat diet CIA rats. Conclusions High fat diet may promote the progress of arthritis in CIA rats by enhancing synovial hyperplasia and inflammatory cytokines.

It is important that we combine animal experiments with clinical medicine though establishment of experimental animals to increase a solid fulcrum for the management of laboratory animals. Also it could greatly promote the development of life science. Therefore, we must provide a complete of animal experiments management system for the good animal experiments' platform. The significance of this study is to improve the related sessions of animal experiment management in medical research center, provide the reference standard of domestic laboratory animal center/room and expand disciplines. The purpose of this study is to provide the practical foundation for the development of disciplines.

    • Comparative Study of the diurnal fluctuations in body weight of mice

      Abstract:Objective To compare the characteristics of diurnal fluctuations in body weight of myopia-induced and normal C57BL6 mice. Methods The total body weight was measured during two consecutive days per 2 hours, and the features of weight changes were plotted and compared. Results With the same age, the weight of experiment group was larger than that of control group (p <0.05). Diurnal fluctuations of weight in both groups arrived an average of 1.3 ~1.6 grams, and appeared as a "sine curve" type without stable point in normal cases, however, the weight of myopia-induced mice was relatively stable in the afternoon. Conclusion The diurnal fluctuations in body weight of mice should attract attention. Myopia induction of C57BL6 mice may affect its characteristics; we recommended evaluating the body weight of experimental mice in a stable period, while normal mice weight measurements should be fixed point in time.

    • Current Status of Mouse Model of Diabetic Nephropathy

      Abstract:Diabetic nephropathy(DN) is one of the main causes of end stage renal diseases. Mice provide a common experimental model of unparalleled flexibility for studying human diseases. Even though some diabetic mouse models exist, not a single one develops renal changes sufficiently reflecting those seen in humans. Progress in identification of effective therapies and pathogenesis for diabetic nephropathy continues to be limited by the lack of ideal animal models. New diabetic mouse models were developed by uses of technique for gene engineering and cross, some of which could recapitulate some typical renal pathological changes of human advanced DN. Here we reviewed the character of common DN mouse models, analyzed disease manifestations, nephrotic features and application of new developed DN mouse models. This would be beneficial for understanding of the disease process and guiding for choosing the most suitable mouse model for the investigation of diabetic nephropathy.

      Abstract:Objective Mamu-B*007:03-sv1 is a splice variant of major histocompatibility complex class I in Chinese rhesus macaque devoid of α3 domain. In this experiment the basic information about expression and cellular location of Mamu-B*007:03-sv1 was got and compared with the full-length Mamu-B*007:03 genes. Methods Eukaryotic expression vectors of Mamu-B*007:03-sv1-myc-pEGFPN3 and Mamu-B*007:03-myc-pEGFPN3 were constructed and transfected to 293T cells. The expressions of the two genes in cells were detected by Western blotting methods, and the cellular locations of the two genes were analyzed by immunofluorescence and laser confocal microscopy. Results Mamu-B*007:03-sv1 and Mamu-B*007:03 were all expressed in the 293T cell and were all glycosylation. While Mamu-B*007:03 gene was expressed on the cell membrane, and Mamu-B*007:03-sv1 gene was expressed in both intra-cellular and cell membrane. Conclusion The expression and cellular localization of Mamu-B*007:03-sv1 deviod of α3 was significantly different from the full length Mamu-B*007:03 gene. Further researches about functions of Mamu-B*007:03-sv1 need to be done.

    • Chronic atlantoaxial catheterization of the spinal subarachnoid space in mice

      Abstract:【Abstract】Objective To establish a new catheterization of the spinal subarachnoid space in mouse and to compare with the traditional catheterization, which can be used for the research on neurophysiology and pain treatment. Methods forty male Kunming mice weighing 24-28g were randomly divided into two groups: Wu group and axis-atlantoaxial group (AA group), respectively catheter from the intervertebral space of lumbar 5-6 and atlantoaxial to the subarachnoid space. After the models were established, we observed the percentage of death and paralysis of limbs, and also measured 50%paw withdrawal threshold (50%PWT), tail flick latency and analgesic effect of morphine to compare the difference of two groups. Results In the case of the same death rate in Wu group and AA group, the percentage of paralysis of limbs in AA group was obviously lower than Wu group. Within 7 days of catheterization, 50%PWT and the TF of AA group was closest to baseline. Morphine analgesic experiment showed the new method adapt to long-term injection. Conclusion The atlantoaxial catheterization model of the spinal subarachnoid space is characterized by its faster recovery and less injury, which was a effective intrathecal catheterization method. 【Key words】Intrathecal Catheterization, Atlas And Atlantoaxial, Animal Model

    • Caloric Restriction Increased Oxidative Damage of SH-SY5Y Cells Activity of Superoxide Dismutase

      Abstract:Objective To detect in the supernatant of superoxide dismutase SOD activity of SH-SY5Y cells, learn about caloric restriction under culture conditions, the cells’ resistance to oxidation. Methods SH-SY5Y cells cultured in vitro, divided into six groups: control(3.0g/L-glucose) group, H2O2(250umol/L)group, low glucose(2.0g/L-glucose) group, low glucose H2O2group, resveratrol(1.25、5umol/L) group, resveratrol(1.25umol/L) H2O2 (250umol/L) group. Determination of superoxide dismutase activity of groups of cells. Results Under conditions of caloric restriction, determination of superoxide dismutase activity: Low glucose and resveratrol(1.25umol/L) pretreat cells for 24 hours, 250umol/L H2O2 treat cell a hour, superoxide dismutase activity showed that, compared with control group, H2O2 group decreased, low glucose group has a significant increase, resveratrol(1.25umol/L) group has no significant change; Compared with H2O2 group, low glucose H2O2 group increased significantly, slight increase in resveratrol H2O2 group. 250umol/L H2O2 treat cell 7 hours, compared with control group, H2O2 group decreased, low glucose group has a significant increase, resveratrol(1.25umol/L) group has no significant change; Compared with H2O2 group, low glucose H2O2 group increased significantly, slight increase in resveratrol H2O2 group. Low glucose H2O2 group significantly higher superoxide dismutase activity of resveratrol H2O2 group. Conclusion Low sugar training method can improve activity of superoxide dismutase in cell culture media, suggesting that caloric restriction by increasing the antioxidant enzymes activity in SH-SY5Y cells, reduce levels of oxidation products, reduce the level of oxidative stress, protects cells from oxidative damage attacks and slow the occurrence and development of related diseases. Resveratrol can’t improve activity of superoxide dismutase in cell culture media against oxidative damage.

    • Analysis of Nutritional Component and Flesh Content of Mugilogobius chulae

      Abstract:Objective To explore the nutrient requirements and compound the feed scientifically for Mugilogobius chulae. Methods The flesh content and nutritional component in the adult fish muscle were measured by conventional methods. Results The flesh content of adult fish were (65.20±3.82) %. Those contents of moisture, crude protein, crude fat and crude ash in the fresh muscle were (79.40±0.03) %, (17.54±0.31) %, (0.62±0.09) %, (2.34±0.07) % , respectively. The limited amino acids were Trp, (Met + Cys). The contents of EPA and DHA in fatty acids of M. chulae were lower. Conclusion Artificial feeds for M. chulae should low the crude fat content and add Trp, Met, Cys, and marine algae such as diatom, brown and red algae.

    • Beijing Region 2011-2012 POLY virus infection of mice Survey and Analysis

      Abstract:[Abstract] Objective to evaluate the prevalence of mice Polyoma Virus in laboratory mice in Beijing by PCR and Serology tests. Method: IFA method was used to test 197 mice sera which were selected from the submitted samples to out monitoring center during 2011-2012. In the 197 samples, 130 sera were from those breeding facilities; and 67 sera were from those experimental facilities. A POLY virus specific primer pair was used in PCR to detect POLY virus from intestines contents of 80 mice which were from breeding facilities. Result: 130 sera from breeding facilities were negative for Polyoma Virus; there were 7/67 POLY virus positive samples from those experimental facilities. A 272bp Polyoma Virus specific amplification can be detected in experimental infected mice. There were no specific amplification in the 80 samples from breeding facilities. Conclusion: the POLY virus pollution were not detectable by the current serological and PCR methods in breeding facilities in the past 2 years: the prevalence of mice Polyoma Virus in experimental facilities can not be neglected. An annual surveillance including POLY virus is necessary not only in productive facilities, but also those experimental facilities.

    • Establishment and Assessment of a Rat Model of Ischemic Reperfusion in Langendorff Perfused Heart

      Abstract:Objective: To explore and summarize the model of ischemic reperfusion in Langendorff perfused heart. Methods:Twenty six Wistar adult rats were divided into two groups randomly: control group and model group, 13 rats in each group. Each rat undergone open-chest surgery, the hearts were removed and immediately canulated and effectively perfused in Langendorff apparatus. Control group hearts only received stabilized perfusion for 105 min and had no ischemia. All the hearts in model group were stabilized for 20 min and then received 30 min ischemia by ligationing LAD( Left anterior descending coronary artery )and 75 min reperfusion。 Evans blue, TTC staining to observe the myocardial infarction area. Compare the level of perfusate CK and LDH ,hemodynamic index , heart rates in various groups at different time:end of 20 min stabilized perfusion, end of 30 min Ischemia,Reperfusion 15 min,30 min and 75 min. Results:According to the Evans blue, TTC staining, myocardial enzymology and hemodynamic indexes, the model group had significant difference compared with the control group. Conclusions: Rat model of ischemic reperfusion in Langendorff perfused heart is stable and effective. This model provides a new way to research on ischemia-reperfusion injury.

    • Improve fertilized ratio of superovulated C57BL/6J by adjusting sperm and egg encounter time

      Abstract:【Abstract】Objective Adjust sperm and egg encounter time to improve the fertilization rate of superovulated C57BL/6J. Method (Ⅰ) To deterimine the fertilization rate by routine superovulated method, which as a control group. (Ⅱ) To verify the insemination ability of C57BL/6J male mouse. (Ⅲ) Determination the time of coitus by check vaginal plug at 3 time points after female and male in one cage, to calculate the time for sperm and egg encounter. (Ⅳ)Female C57BL/6J mouse after superovulatory treatment, stayed for 6.5 h before matinge with male mouse. Collected fertilized eggs,calculated the rate of fertilization and compared effects between method changed before and after. Result (Ⅰ) 10.88 fertilized eggs were collected for one mouse in control group; (Ⅱ) Average 20.55 fertilized eggs were collected in the second group, increased 88.87% than control group, that demonstrated the ability of male mouse for getting a high fertilization rate in natural, and the encounter time for sperm and egg was the key factor on fertilization rate; (Ⅲ) For majority mice with plugs at 4 h after cohabitation, the sperm and egg encounter time in control group could be assured that was approximately 7 h; (Ⅳ) Based on the results of above, delayed 6 h for female and male mice feeding together, the mean number of fertilized eggs for one mouse was 22.27, raised 105% than control group. Conclusion Adjusting the sperm and egg encounter time can significantly increase fertilizer ratio of superovulated C57BL/6J mouse.

    • The Comparison Study between Repeated Cryo-injury and Coronary Artery Ligation Model of Rats

      Abstract:【Abstrct】Objective To explore the two different methods of repeated cryo-injury and coronary artery ligation to establish rats model of acute myocardial infarction(AMI),and to compare the rats’ mortality and parameters of cardiac function.Methods: Forty SD rats (males 20) were randomized into two groups:cryo-injury group(CIP)(n=20)and coronary artery ligation group (CAL)(n=20). Cryo-injury of the rats’ left ventricle free wall was performed with the use of a round copper probe cooled by immersion in liquid nitrogen without ventilator support. It was applied to the LV free wall three times for each five seconds. The CAL group was ligated anterior descending coronary with ventilator support. We observed the rats’ mortality during operation, post-operation period and 28-day. Parameters of cardiac function were observed at the 28-day(SAP、DAP、LVEDP、LVMI、眃p/dtmax). Results ①The death number of rats in CIP group has no statistical significance in intraoperative、postoperative、28 days compared with the CAL group(P>0.05); Compared with CAL group,the rats total mortality of CIP group was obviously decreased(P<0.05).. Rats death amount of CIP group was obviously less than CAL group; ②At 28-day,in CIP and CAL groups,the cardiac function parameters (SAP、DAP、LVEDP and LVMI)were increased and 眃p/dtmax was decreased,The result showed two groups of rats cardiac function deterioration.There was no statistically significance in cardiac function (P>0.05).Conclusions: Repeated cryo-injury method deserves recommendation due to the advantages, such as safety and low mortality without ventilator support to establish rat model of acute myocardial infarction.
