Abstract:【Abstract】 Objectives The NAD-dependent deacetylase SIRT1 appears to have evolved complex roles in central nervous system. Recent studies suggest that SIRT1 has been involved in the processes of glucose and insulin secretion. The aim of the present study is to investigate if SIRT1 co-locates with IR in the adult C57BL/6 mice brain, which may provide morphological evidence for inherent relationship between SIRT1 and IR in brain. Methods Using double immunofluorescence staining to investigate if SIRT1 co-locates with IR and the distribution in the brain. Results SIRT1 immunoreactivity was mainly observed in the hippocampal CA1 pyramidal cells. The co-location analysis showed that SIRT1 was co-located with IR in the hippocampus, cortex and hypothalamus of adult mouse brain. In vitro, insulin regulated the expression of SIRT1. Conclusions SIRT1 was rich in neuron and collocated with IR. The results provide a morphological clue to explain the role of SIRT1 and IR in the brain function and neurological disorders.

Abstract:Tree shrew, a kind of lower-grade primates animals, whose value at biology research has cause attention of researchers, have been more and more demanded as laboratory animals. However, as the south subtropical animals, how to adapt to life in the northern area, is the premise that tree shrew can be used as laboratory animals in northern area. Our laboratory has explored feeding cage, which is suitable for Tree shrew living; detemined feeding environment, such as temperature, humidity, illumination time, noise, ventilation and so on; formulated the standardized health management standard and established breeding management method of long-term artificial cultivation of Tree shrew; which have lay the foundation for research on traditional Chinese medicine syndrome combined with disease animal models using Tree shrew in the future.

Abstract:To investigate the effect of oral administration and medicine bath of BuYangHuanWu decoction on the Sciatic Nerve Conduction Velocity(SNCV). Method: 60 Sprague Dawley rats whose left sympathectic ganglion were exposed were divided into two groups. The rats in the experimental group take oral administration and medicine bath of BuYangHuanWu decoction . The SNCV were measured both before and after operation. Results:The SNCV in the experimental Group were significantly faster than those in the control group A(p<0.01) after treatment for 2,4,6 weeks. Conclusion: It is beneficial to improve the SNCV by oral administration and medicine bath of BuYangHuanWu decoction.

Abstract:Systemic lupus erythematosus(SLE) is characterized by the presence of anti-nuclear antibodies (ANA) directed against naked DNA and entire nucleosomes. Systemic lupus erythematosus is a multifactorial autoimmune disease that affected by genetic, environmental, hormonal, infection factors. But mice with lupus nephritis model and SLE pathogenesis and pathological changes have similar characteristics to human in SLE, which is an ideal experimental pathological model for the study of SLE and screening of anti-inflammatory-immunity drugs. In this article we have reviewed lupus nephritis mouse model preparation methods and evaluations. This would be of its significance through the comparison of murine lupus nephritis model specific methods and evaluation of characteristics.

Abstract:Objective To measure IL-1β, IL-1R1, IL-8, IL-8R and IL-15 genes expression in rabbit sacculus rotundus using the Liquid suspension array system, and evaluate the method. Methods The simultaneous measurement and quantity of multiple genes mRNA expression in rabbit sacculus rotundus was detected by the Panomics QuantiGene Plex 2.0 Assay of Affymetrix using the bDNA signal amplification and beads analysis techniques. Then, this method was applied in the detection of rabbit interleukin gene. Results Content of IL-1β, IL-1R1, IL-8, IL-8RA and IL-15 gene can be detected simultaneously. Gene expression of IL-15 in WHBE rabbit is significantly higher than JW rabbit (P<0.05), and gene expression of IL-1R1 in WHBE rabbit is significantly higher than JW rabbit (P<0.01). Also, the relative expression quantity of IL-8RA gene in WHBE rabbit is higher than JW rabbit (P<0.05). Conclusion Liquid suspension array method for detecting rabbit interleukin gene was established. Gene expressions of IL-15, IL-1R1 and IL-8RA in WHBE rabbit were higher, and all the results reveal WHBE rabbit may have unique immunological properties.

Abstract:Objective To observe the change of Neuroendocrine hormone and Electrolyte levels in WHBE Rabbit with Irritable Bowel Syndrome, to explore the regulation role of nerve-humoral-endocrine system. Methods The irrtable bowel syndrome rabbits model was made by using the methods of moist heat, stress and stimulation adding with low-dose induced cathartic induced. When modeling 9d and 14d, take blood samples, testing the changes of rabbits serum levels of dopamine (DA), 5-hydroxytryptamine (5-HT), adrenocorticotropic hormone (ACTH), cortisol (COR), β-endorphin (β-EP) hormone and K , Na , Cl- content, and compared with JW rabbit. Results Compared with the normal control group, model 9 days group WHBE rabbits DA, 5-HT, ACTH, β-EP and COR significantly increased (P<0.05, P<0.01), Na , K content were significant with lower (P<0.01), while the JW rabbits ACTH, β-EP and cortisol are significant with increased (P<0.05, P<0.01), K content significantly with lower (P<0.05); with the normal control group ratio, model 14 days group WHBE rabbit 5-HT, ACTH, β-EP were significantly increased (P<0.05, P<0.01), JW rabbits ACTH significantly increased (P<0.05). The correlation analysis showed, IBS model group serum 5-HT, COR and Na , K concentration showed a significant negative correlation (P<0.05, P<0.01), serum DA, β-EP level and Na , K , Cl-concentration was a significant negative correlation (P<0.05, P<0.01), ACTH levels with K concentration was significantly negatively correlated (P<0.01). Conclusion IBS is not only associated with elevated HPA axis excitability, but also humoral factors, nerve-humoral- endocrine system is involved in the occurrence of WHBE rabbit with IBS.

Abstract:【Abstract】 Aim To compare the success rate and the incidence of complications together with the time spent of two kinds of femoral artery punctures methods in the preparation of acute myocardial infarction mode.Methods Sixteen mini-pigs were randomly divided into two groups:the ultrasound-guided puncture group (n=8) and the puncture group under direct vision by cutting(n=8).Th e related success rate and the incidence of complications together with the time spent of both groups were investigated.Results The success rate and incidence of infection in two goups are 100% and 0% respectively.Major bleeding complications in the puncture group under direct vision by cutting was lower significantly compared with the control group(P<0.O5). The time spent were also lower significantly compared with the control group(P<0.05).Conclusion The femoral artery punctures methods under direct vision by cutting is a safe and efective method, also a relatively time saving method.

Abstract:Objective: To study the effect of BRL-37344 on alveolar fluid clearance (AFC) in isolated human lung and the mechanisms involved. Methods: Isomolar albumin solution in the presence of 10-4mol/L to 10-8mol/L BRL-37344 (selective β3-adrenergic agonists) was injected into the alveolar spaces in isolated human lung, and AFC was measured. 10-5mol/L BRL-37344 combined with phentolamine (α receptor antagonist), atenolol (β1 receptor antagonist), ICI-118551 (β2 receptor antagonist), SR59230A(β3 receptor antagonist), sodium channel blockers or Na /K _ ATPase blockers were perfused into the alveolar space of isolated human lung. Results: 10-4mol/L to 10-7mol/L BRL-37344 increased AFC significantly in isolated human lung,but 10-8mol/L BRL-37344 can not increase AFC. The AFC-stiulating effect of BRL-37344 was lower with SR59230A, amiloride and ouabain. Conclusion: BRL-37344, aβ3-adrenergic agonist, can increase AFC in isolated human lung through promoting the transport of Na -K -ATPase and accelerate the amelioration of pulmory edema.

Abstract:Purpose: Research on the applied result of the multifunctional integrated tubular pipeline in femoral artery and vein in the animal experiment. Methods, in 31 severe multiple wounds with self-made multifunctional integrated tubular pipeline in femoral artery and vein, observe the time and effect on blood transfusion, fluid infusion, blood sampling, administration, measurement of central venous pressure on sequence of all process. Result. The average time cost in the animal experiment with the multifunctional integrated tubular pipeline is 80 /-12 seconds. The average time cost on each item is 19 /-5.1 seconds. Conclusion: The multifunctional integrated tubular pipeline in femoral artery and vein saves time and human resources and works quickly and conveniently in animal experiment, so it is worthy a further applied development.

Abstract:Objective Rat is a common animal for making heart disease model. The magnetic resonance imaging (MRI) is becoming an important technology for study of pathological process and drug evaluation with animal model. However, no normal cardiac MRI technological parameters of rats are available at home and aboard, and limit this technology application. In this work, the technological parameters for structure and function of left and right ventricle from normal adult rats of Wistar, Sprague-Dawley and Lewis were acquired and analyzed quantitatively by magnetic resonance imaging (MRI), and provided as reference data for heart disease model preparation and analysis. Methods The cardiac CINE was used in a 7.0T high field MRI to assay the structure and function parameters of left and right ventricle of the rats. Results The structure and function parameters of left and right ventricle of the rats were acquired and analyzed, including left and right ventricle end-diastolic volume (EDV), end-systolic volume (ESV), ejection fraction (EF); left ventricle papillary muscle lay end-diastolic diameter (EDD), end-systolic diameter (ESD), fractional shortening (FS), end-diastolic anterior wall thickness (EDAWT), end-diastolic posterior wall thickness (EDPWT), end-systolic anterior wall thickness (ESAWT), end-systolic posterior wall thickness (ESPWT), anterior wall thickening (AWT) and posterior wall thickening (PWT), cover right ventricle papillary muscle lay end-diastolic wall thickness (EDWT), end-systolic wall thickness (ESWT) and wall thickening (WT). Conclusion This study obtained quantity information of 18 cardiac structure and function parameters of three normal rat lines, which can be used as reference data for heart disease model preparation, pathological process and drug evaluation.

Abstract:Objective To discuss the change of pathology and biochemical indexs during the building the Mini-pig model of hepatic cirrhosis. Method The subjects were 50 Bama mini-pigs of the same germline, who were divided at random into a experiment group of 40 and a control group of 10, male or female, aged 4 months, weighing 15-20 kg. Composite factors were employed to establish a model for the experiment group, and the normal feeding was synchronically given to the controlled. At 0th、4th 、8th、12th、16th、20th、24thWeekend,the liver puncturation biopsy and the haemospasia by the ear venous were done. Result At 24thWeekend, the normal group was 70 cases, the slight hepatic fibrosis(stage S1+S2) was 82 cases, the severe hepatic fibrosis(stage S3+S4)was 62 cases, and the hepatic cirrhosis was 33cases. From normal to cirrhosis groups, ALT、AST、T.BIL、CR and GLO of the pathological models rose gradually, ALB decreased gradually and A/G ratio was inversion, PT prolonged gradually, there were statistically significant in groups(p<0.01). Conclusion The model-building from composite factor food of carbon tetrachloride (CCL4) can establish successfully the models of Mini-pigs liver cirrhosis. The pathology and biochemical indexs change obviously in different stage of hepatic fibrosis.

Abstract:Objective To investigate the B virus infection rates in rhesus macaques changing with seasons. Methods B virus antigen ELISA kit was used to detect B virus antibody of 54 weanling rhesus macaques for 13 consecutive months. Results In December, January, February, March and April, the number of B virus positive monkey is increased significantly, and in other months the number of B virus positive monkey is relatively stable. Conclusion B virus infection is affected by the seasons, in spring B virus infection rates is higher, that can strengthen the precautions or regulate environmental factors in high seasons to reduce B virus infection and improve the monkeys quality.

Abstract:The present study was undertaken to clarify the effect of the first generation histamine H1 receptor antagonists on the adenosine triphosphate-sensitive potassium (KATP) channel in the isolated mouse ventricular cardiomyocytes using excised inside-out and cell-attached patch clamp techniques. In the excised inside-out patch configuration, H1-antihistaminic agents (chlorpheniramine, pyrilamine and diphenhydramine), doses ranging from 1 to 100 μmol/L, inhibited KATP channel activity in a dose-dependent manner. The potency order reducing the channel activity was pyrilamine > diphenhydramine > chlorpheniramine. All the antihistamines (100 μmol/L) also inhibited the pinacidil-induced KATP channel activity in the cell-attached patch configuration. The potency order of the 3 antihistamines inhibiting KATP channel activity is pyrilamine > chlorpheniramine > diphenhydramine in the cell-attached configurations. Histamine did not affect the pinacidil-induced KATP channel activities by itself, in addition, did not influence the effects elicited by the 3 antihistamines on the pinacidil-induced KATP channel acitivities in the cell-attached patches. From these results, it concluded that the first generation histamine H1 receptor antagonists are involved in the regulation of the ATP-sensitive potassium channel activities in the mouse cardiac ventricular myocytes, and that the inhibitory actions of the antihistaminic agents on the channels are not dependent on the H1-receptors.

Abstract:Objective Use the special and double immunohistochemistry staining method to obsearve the typical histopathological change in experimental acaques diabetes model.Methods we used STZ (30 mg/kg of body weight, intravenously, 3days)to five macaques. The animal was sacrificed when impending death. The histopathological variety of pancreas,heart,kidney and spleen were observed with H.E,PAS,Masson,Sirius red,toluidine blue and double immunohistochemistry staining. ResultsThe quantity of pancreatic island decreaseded obviously,the reminders showed atrophy. fiber hyperplasia were observd in exocrine pancreaswith masson stain. In kidney, There were focal chronic nephritis, renal interstitial fibre hyperplasia, parts of glomerular and renal tubular atrophy.Small arteria in spleen was thickening with PAS stain. In double immunohistochemistry staining, the quantity of brown-colored A cell is increased while the pink-colored B cell is decreased. Concusion The use of H.E combined with special and double immunohistochemistry staining is a better method to evaluate the Diabetes Mellitus model.

Abstract:Objective To search for some key genes which account for the different sequelae(chronic woodchuck hepatitis and acute self-limiting woodchuck hepatitis) of woodchuck hepatitis virus infection between neonatal and adult woodchucks. Methods Applying the genome-wide expression microarray, some related genes were selected. With the degenerate primers designed on the iCODEHOP from clustal of some different species, some products were amplified from the woodchuck cDNAs. After the products sequencing done, some accurately designed primers generated from the Primer Premier 5.0, Real time quantitative PCR performed ultimately. Results Compare to the neonatal woodchuck, calcium reabsorption related gene, in the adult woodchuck liver cells DNM1 ( Dynamin 1 ), DNM3 ( Dynamin 3 ) and Prkcc ( Protein kinase C, gamma ) expression rate increased significantly, by 2.65?0.25 times, 1.90 ?.34 times, 2.94 ?.54 times. Conclusion In the endocrine and other factor-regulated calcium reabsorption pathway, these three genes have the most significant changes in mice, as well as in woodchucks at the two time points. Those genes make contribute to the differential calcium concentration, and promote woodchuck hepatitis virus replication indirectly. In a word, these genes could have an effect on the sequelae of woodchuck hepatitis virus infection at different ages.

Abstract:Objective To develop a RT-PCR method for determination of Mouse Hepatitis Virus in Laboratory mice and Mongolian gerbils. Methods Two pair of primers specific to the sequences of S gene were designed according to the published Mouse Hepatitis Virus.With which the RNA of MHV was extracted and reversely transcribed to cDNA as a template for PCR amplification. The developed RT-PCR method was optimized, carries on the specificity, sensitivity, stability, repeatability test. And used for determination of 65 Mongolian gerbils and 12 mice.Results The developed RT-PCR method is good in specificity, ensitivity, stability, and repeatability; and its minimum detection limit using the recombinant plasmid containing MHV gene as atemplate was 3.1pg/μl,and the lowest detection virus titer is 10-3.ml-1.The 65 Mongolian gerbils after RT - PCR detection were negative;The 12 mice after RT - PCR detection, there were three MHV positive, compared with the MHV in Genebank, The homologies in nucleotide sequence of 3 positive mice all is 97%. Conclusion The developed RT-PCR method can be used in detecting the Mammalian Mouse HepatitisVirus (MHV) in laboratory animal ,such as mice and Mongolian gerbils.
