Abstract:Objective Measure and compare the body weight, body length and chest circumference of SPF KM, NIH and BALB/c mice. Methods We mated KM, NIH, BALB/c six times, when we got offspring, and measured them at 1d, 14d, 21d, 28d, 35d, 42d, 49d, 56d and 63d. The weights were tested at electronic balance and the body length and chest circumference were tested by ruler. Result and Conclusion KM mice were similar to NIH mice in the body weight, body length and chest circumference. But BALB/c mice had lots of differences from both of the KM and NIH mice.

Abstract:The characters of rat on physiology, behavior and metabolism are more similar with human being compared with that of mouse. With the development of rat genetic engineering techniques in recent years, the genetic modified rats have been a hot point of laboratory animal science and back to the life science and pharmaceutical research. The advantages and disadvantages of the transgene, transposon, ZFN (zinc finger enzymes), TALENs and embryonic stem cell-based gene targeting were compared in this review paper.

Abstract:【Abstract】 Objective To investigate more simple and effective method of rat orotracheal intubation. Methods 40 rats were randomly divided into two groups. The intubation time and intubation frequency, the success rate of the first intubation, the survival rate in 1 week were compared between the improved group and the conventional group. Results The time and frequency of intubation were significantly less in the improved group than those in the conventional group[(10.83?.69) s vs (36.41?.71) s,1.04?.21 vs 1.78?.03,p<0.05].The success rate of the first intubation in the improved group was obviously higher than that of the conventional group(95% vs 60%, p<0.05).There was no obviously difference in the survival rate after extubation in 1 week between groups (100% vs 85%, P>0.05).Conclusion The improved method is superior to the conventional method, Homemade instruments is worth promoting.

Abstract:Objective Investigate the effects of salvianolic acid A on rats of bleomycin-induced pulmonary fibrosis and potential mechanisms. Methods Single-dose bleomycin was given via intratracheal on Day 0 and rats were treated from Day1 daily with tail-vein injection of salvianolic acid A for 28 d. Record weights and observe general physical sign. Measure free radical、pulmonary function and take electron microscopes. Results Salvianolic acid A could recover physical sign, enhance SOD and MDA activities and reduce MDA contents. Salvianolic acid A could minimize of alveolar spaces and alleviate the inflammation. Moreover, it could improve pulmonary function and decrease collagen expression, inhibits type Ⅱ alveolar epithelial cells hyperplasia in pulmonary fibrosis rats. Conclusion Salvianolic acid A could attenuate the bleomycin induced pulmonary fibrosis of rats and potential mechanism may be correlated with antioxidation of SAA.

Abstract:Diabetes is a kind of endocrine metabolic disease, which mainly including type 1 diabetes and type 2 diabetes, most of them are the latter, with low course and different degrees of complications. The incidence rate, disability rate and fatality rate of diabetes are quite high, which bring a big challenge for the treatment. For now, there are many researches and discussions in animal models of type 2 diabetes complications at home and abroad, mainly through the injection of STZ to induce type 2 diabetes complications, which can show the lesion characteristics better, all of these study can provide some references for the prevention and curing of type 2 diabetes.

Abstract:Objective: fMRI animal experiments that require sustained anesthesia to provide a simple, convenient, and safe manipulation condition. This investigation was designed to confirm the appropriate anesthetic regimen for fMRI experiment of rhesus. Method: Seven rhesus from Academy of Military Medical Sciences are female, weight 4-7 kg using continuous intravenous anesthesia (ketamine propofol) . Ketamine 10mg/kg im until the rhesus sleep then establish a continuous intravenous access. continuous injected ketamine100mg/20ml, and propofol 3mg / kg / min, then intermittent ketamine 1mg/kg/30min. The rhesus placed in a special experiment rack and the incisor hold up to keep the airway open. During the whole test we monitored breathing, heart rate, corneal reflex and pupil. Results: 44 times experiments successfully completed, no animal restlessness, respiratory depression, arrhythmia, deaths and other serious complications. At the end of the experiment, the experimental animals awake soon. Conclusion: continuous intravenous anesthetic of propofol and ketamine could provide more secure, convenient and practical method for fMRI animal experiments.

Abstract:【Abstract】 Objective To observe the anesthetic effect of ketamine combined with alcohol on experimental dogs in state of swaying naval vessels. Methods 34 dogs were divided into group Ⅰ(n=10,in state of static naval vessels) and group Ⅱ(n=24,in state of swaying naval vessels). The dog in two groups was anesthetized by ketamine respectively, then alcohol was given by intravenous drip. The anesthetic effect was recorded. Results The induction time of basal anesthesia for ketamin in group Ⅰwas no significant difference than that in group Ⅱ(P﹥0.05).The anesthesia of ketamine can ensure the accomplishment of skin preparation and placement of venous trocar for dogs, and the anesthesia can be maintained with alcohol continuous infusion.Anesthetic effect was divided into good,moderate,bad,and death according to the reaction of dogs,and the anesthetic effect rate(good+moderate)were 100% in two groups. Conclusions The anesthetic effect of ketamine combined with alcohol on dogs is safe,effective for the anesthesia on experimental dogs in state of swaying naval vessels.

Abstract:Objective: isolation and culture of Tibetan miniature pig rib chondrocytes (PCCs), followed by identifying it. Methods: a 3-day-old Tibetan miniature pig was used in this experiment. Both PCCs and mouse rib chondrocytes (MCCs) were isolated by two-step digestion method. The isolated chondrocytes were identified by toluidine blue staining and immunofluorescence for collagen II and aggrecan. Tibetan miniature pig embryonic fibroblasts (PEFs) and mouse embryonic fibroblasts (MEFs) were employed as a negative control. Results: PCCs and MCCs were successfully isolated and cultured, and demonstrated the different cell morphology, whereas toluidine blue staining and immunofluorescence staining confirmed the chondrocytes characteristics of isolated PCCs. Conclusion: The method for isolating, culturing and identifying PCCs of Tibetan miniature pig was successfully established.

It is an effective means to research human aging process by using natural aging animal or anthropogenic aging animal as a model. Abstract:To establish animal models of liver tumors, the PLCε gene knockout mice was induced by mutation inducers Diethylnitrosamine (diethylnitrosamine DEN) combined with tumor enhancer phenobarbital (phenobarbital PB). Methods 12-day-old male PLCε-/ - and PLCε + / + mice were randomly selected as group I (n= 40)and group II (n= 40),respectively.The mice of group I and II were injected DEN through abdominal cavity at the age of 12 days,then drinking PB after 4 weeks and continuously until 24 weeks. At the same time, 12-day-old male PLCε-/ - and PLCε + / + mice were randomly selected as the control group I (n= 40) and control group II (n= 40), respectively, and fed with normal feeds for 24 weeks. At the end of the experiment, the liver tumor was observed under electron microscopy. Results Pathology results confirm that 18 mice of group II had liver tumors, and rate of tumor formation is 60%; 15 mice of group I had liver tumors, and rate of tumor formation is 46.9%; the mice of control group I and II had not liver tumors. Conclusions The animal model of liver tumor have been successfully established by using mutation inducers DEN combined with PB in PLCε gene knockout mice, and could provide a foundation for further researching effects of PLCε during liver tumor formation.

Abstract:Objective To evaluate the influence of Xuebijing (XBJ) by inflammation cytokines of multiple organ dysfunction syndrome (MODS). Methods 90 rats were randomly separated into 3 groups, the control group and XBJ group were infused 15 μg / kg LPS in vena caudalis, Then the physical sign appearance was observed and collection the serum to determine the expression concentration of TNF-α、IL-1α、IL-4 and IL-10 by ELISA in each group at different times. Results ① The death rate of rats were the percent of 0, 37, and 20 in the control group,model group and XBJ group, respectively. ② The XBJ enhance the increased amplitude of TNF-α and deduce the increased amplitude of IL-1α by compared with model group, the expression concentration peak of TNF-α and IL-1α is 6h~24h. ③ The expression concentration of IL-4 in model group and XBJ group in model group has significant ascendant compared with control group at 6h~48h; XBJ group has significant have significant ascendant compared with control group at 6h~12h (p<0.05).The expression concentration of IL-10 ascendant compared with control group at 6h (p<0.05). Conclusion The results demonstrated that XBJ could degrade the death rate of MODS induced by LPS,and it could reduce the increased amplitude of TNF-α, whereas enhance the increased amplitude of the serum IL-1α,IL-4 and IL-10, it could also prevent those rats from changing into MODS efficiently.

Abstract:[Abstract] Object: To investigate effects of rearing density on growth and development of the zebrafish. Materials and method: Zebrafish was maintained at 1, 2, 4, 6, 8 Tail/L density in a 3L indepent unit. In some distinct developmental stages, the weight, length, organ weight of fish was measured and some indicators were calculated. The results of weight, DWG, GBW, GBL and viscera index were analyzed by SPSS 17.0. Result: There was no significant difference among each group ,when the rearing density differ from 1 to 4 Tail / L. However, the weight, DWG, GBW, GBL and viscera index descend when density is increasing, and there was a significant difference between the groups of low density (P< 0.01). Conclusion: In a comprehensive consideration of considering the factors that affect the efficacy of breeding including space and the quality of living environment of zebrafish et al , 4 Tail / L is an ideal rearing density.

Abstract:Objective To study the polymorphism and mechanism of GDF-9 and BMP-15 in mice of different strains, different reproductive capacity, and find out the relationship between genes and fecundity. Methods Screening polymorphism loci by using direct sequencing method, and testing 5 'UTR impact on downstream genes by constructing luciferase dual reporter vector. Results ICR mice litter size is about 2.8 times the BALB/c; In ICR and BALB/c, respectively, found three polymorphisms, one at the 5 'end of the non-coding region, the other two located in the protein coding region ; Dual-luciferase test results show that the 5'UTR area of GDF-9 can promote the expression of the downstream gene. Conclusions GDF-9 polymorphic loci exist in ICR and BALB/c strains, changes in these sites play a certain role in promoting the expression of the downstream gene.

Methods: Through the ten successful implementation of the LVAD on goats, considering with the characteristics of the animals and in accordance with the requirements of the regulations on the administration of experimental animals, the managements were formulated on aspects of the perioperative environmental control and postoperative special management to ensure the normal operation. Results: Through the science rigorous management practice, succeeds to implement 10 cases of goat implantation type left heart auxiliary unit (LVAD) surgeries, guarantees the normal operation of surgery.

Abstract:Objective: To estimate the genetic diversity of Macaca mulatta and providing valuable information and references for standardization of the population. Methods:Macaca mulatta were genotyped using 19 microsatellite DNA markers, DNA were extracted from blood and amplified by PCR, PCR products were analyzed by polyacrylamide gel electrophoresis. Every microsatellite locus was calculated by softwrare Popgene 1.32 and Excel. Results: All nineteen microsatellite loci were highly polymorphic, 164 alleles were found in the outbred stock. Na, Ne, Ho, He, PIC and I ranged from 6 to 11, 3.5727 to 8.9416, 0.2560 to 0.6364, 0.7230 to 0.9010, 0.6771 to 0.8874, and 1.4249 to 2.2662, with an average of 8.6316, 6.0709, 0.4309, 0.8270, 0.7979 and 1.8901, respectively. All of 19 loci showed significant deviations from Hardy-Weinberg equilibrium. Conclusions: Results indicated there are high variations of Macaca mulatta in this study. The experiments can provide a theoretical foundation for establishing genetic quality monitoring method with specific microsatellite loci.

Abstract:Objective To get recombinant antigen including the antigen epitope of gB protein for the detection of B virus. Methods The gene fragment of the antigen epitope sequences of gB protein was synthesized and directly cloned into the pET-22b vector. The positive recombinant vector was transformed into BL21 competent cells and induced with IPTG. Results The synthesis gene fragment was expressed successfully in the Prokaryotic cells on the soluble form. Conclusion Prokaryotic expression system can produce soluble B virus gB protein antigen and can be used as the detection of B virus antigen
