Abstract:Objective This study was designed to determine the effect of transient receptor potential vanilloid type 4 (TRPV4) on angiotensin II (Ang II)-induced renal injury in TRPV4-null mutant (TRPV4 - / - ) mice. Methods The mice were divided into sham group and Ang II-treated group. Ang II was infused systemically into wild type (WT) and TRPV4 - / - mice via a miniosmotic pump for 4 weeks, and the sham mice were given with normal saline. Systolic blood pressure, urinary excretion of albumin and 8-isoprostane, serum creatinine, and the pathological changes in the kidney tissues were assayed after the 4-week treatment. Results Compared with corresponding sham mice, Ang II infusion led to enhanced systolic blood pressure, increased urinary excretion of albumin and 8-isoprostane, increased serum creatinine (P< 0.05), and enhanced glomerulosclerosis degree and renal tubulointerstitial injury index (P < 0.05) in the WT and TRPV4 - / - mice. The result were associated with enhanced collagen levels in the kidney (P < 0.05). All of them were attenuated by the deletion of TRPV4 in the absence of alteration in blood pressure (P < 0.05). Conclusions Deletion of TRPV4 could alleviate renal injury during Ang II-induced hypertension, suggesting that TRPV4 may contribute to the pathophysiology of angiotensin II-induced renal injury.

Abstract:Objective To study the protective effect of Panax notoginseng saponins (PNS) and its components Rg1 and Rb1 on oxygen-glucose deprivation/ reoxygenation ( OGD/ Reox)-induced tight junction damage. Methods Anaerobic box were used to induce OGD in HUVEC cells for 6 h followed by reoxygenation for 24 h. Transepithelial/endothelial electrical resistance (TEER) and cell permeability were detected, immunefluorescence was used to observe the ZO-1 and claudin-5 protein expression. Results PNS 20, 40 mg/ L and ginsenoside Rb1 significantly inhibited the OGD/Reox-induced decreased tight junction resistance, and the increased cell permeability (P < 0.05). PNS 20, 40 mg/ L and ginsenoside Rb1 partly restored the inter-cellular tight junctions which were regularly arranged on the cell membrane, and the cells displayed cobble stone-like arrangement. Conclusions PNS ameliorates ischemia-induced vascular endothelial cell tight junction damage via MMP-9 and VEGF/ VEGFR2 signaling pathway. Rb1 is one of the effective monomer components.

Abstract:Objective To investigate the effect of recombinant botulinum neurotoxin serotype A heavy chain (BoNT/ A heavy chain) on local proteins which are related to nerve growth after spinal cord injury in rats, and to get some experimental evidence to explain the mechanism of BoNT/ A heavy chain in stimulating neuritogenesis. Methods Recombinant botulinum neurotoxin serotype A heavy chain was applied locally or intrathecally to rats with ipsilateral semidissociated lumbar spinal injury. Local spinal tissue was extracted for general protein expression by two dimension electrophoresis plus nitrate silver staining after different time period of injury. Based on the results of 2-D gelelectrophoresis, growth-associated protein 43 (GAP-43) and of superior cervical ganglion 10 (SCG 10) were selected to examine the changes of their expression and distribution features under BoNT/ A heavy chain administration using SDS-PAGE, western blot and immunofluorescence. Results (1) The model of spinal cord injury (SCI) in this study was an ipsilateral semi?dissociated lumbar SCI in rat. The rats showed obvious motor and sensory dysfunction in the ipsilateral hind limb. (2) The results from 2-D gel electrophoresis plus nitrate silver staining showed that the administration of BoNT/ A heavy chain based on SCI altered the local protein expression pattern. The decrease or increase in the expression of some protein dots / dots group was clearly seen after single SCI. However, these changes were transformed by BoNT/ A heavy chain treatment, which appeared as a reversed pattern turning toward that in control group or further increased expression upon SCI, such as the dots located respectively at 35 -45 kDa and 18 -25 kDa level, pI between 5 -7. In addition, the expression of the two dots located at the level as above increased after SCI only, and showed further increase in their expression with BoNT/ A heavy chain intervention. (3) The changes of selective GAP-43 and SCG 10 expression and distribution by western blot and immunofluorescence indicated that the administration of BONT/ A heavy chain based on SCI amplified the expression of GAP-43 and SCG 10 (P < 0.05). Meanwhile, the positive immuonfluorescent staining for both GAP-43 and SCG 10 mainly distributed nearby the proximal area of injury, both cytoplasm and neuronal processes were positively stained. Conclusions Intrathecal delivery of BoNT/ A heavy chain increases the expression of growth-associated proteins GAP 43 and SCG 10 after SCI in rats.

Abstract:Objective To investigate the effect of vitamin D (VD) deficiency on the pathogenesis of Zucker diabetic fatty (ZDF) rats. Methods Male 5 -6 weeks old Zucker rats were randomly divided into 4 groups according to their body weight: normal control group (ZL), VD deficient control group (ZL + VD. Def), model group (ZDF) and VD deficient model group (ZDF + VD. Def). All the rats were fed to 12 weeks of age, and body weight, food intake, water intake, urine volume, urine glucose, fasting blood glucose were measured. The glucose tolerance was tested at 11 weeks. Pancreatic samples were taken and tissue sections were examined by pathology using HE staining. Results The body weight of ZDF + VD. Def group was higher than that of the ZDF group. Drinking water and urine volume were increased earlier than the ZDF rats. The blood glucose in the ZDF + VD. Def group was increased significantly earlier than ZDF group, it is about 2 times of the ZDF group at 12 weeks. Compared with the ZDF group, the impaired glucose tolerance and islet damage were more serious than the ZDF + VD. Def group. Conclusions Vitamin D deficiency accelerates and aggravates the pathogenesis of ZDF rats, and VD deficiency may be a key factor in the pathogenesis of obese DM Type 2.

Abstract:Objective To study the hematopoietic stem cell injury(HSC) induced by busulfan. Methods C57BL/6 mice were treated with i. p. injection of 20 mg/ kg or 40 mg/ kg busulfan. All mice were euthanized at 15 days and 30 days after busulfan treatment for analysis of the peripheral blood cell counts, bone marrow cellularity and HPC (LKS-, lineage-sca1- c-kit+ ), HSC (LSK, lineage-sca1+ c-kit+ ) and long-term HSC (CD34- LSK, CD34- lineage-sca1 + c-kit+ ) frequency. The colony-forming unit-granulocyte and macrophage (CFU-GM) ability of HPC was measured by colony-forming cell (CFC) assay, and the HSC self-renewal capacity was analyzed by single-cell colony-forming assay. Results The busulfan administration decreased the WBC, RBC and PLT compared with control mice. The HPC function (CFU-GM) was impaired (P < 0.05), and the HSC colony forming ability was decreased at 15 days after busulfan treatment (P < 0.05), whereas the body weight of the mice didn’ t change significantly after busulfan treatment. Conclusions Our findings suggest that busulfan can induce hematopoietic stem cell injury, and provide a support for the study of hematopoietic stem cell injury mechanism.

Abstract:Objective To develop an ideal hypertension combined hyperlipidemia (HP/ HL) rat model by feeding spontaneously hypertensive rats (SHRs) with high fat diet, and to evaluate the pathological changes in target organs including heart and kidney. Methods Twenty 3-week old male SHRs were randomly divided into two groups: normal fat control group (SHR-NC) and high fat group (SHR-HF). Moreover, ten 3-week old male Wistar-Kyoto rats (WKY) were taken as the model control group (WKY-NC). The rats in SHR?HF group were fed with high?fat diet to induce HP/ HL, while rats of WKY-NC and SHR-NC groups were fed with normal diet. The systolic blood pressure (SBP) and body weight were measured every week. At the end of the experiment, the rats were sacrificed to take serum samples for blood lipid analysis including high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), total cholesterol (TC) and triglyceride (TG). Heart and kidney tissue samples were collected to examine the pathological changes using HE and Masson staining. Results Compared with the SHR?NC group, the SHRs fed with high-fat diet for 23 weeks presented significant increase of blood pressure and TC, TG, LDL-C, and decrease of HDL-C. The HP/ HL rat model showed pathological changes in the HP/ HL target organs, heart and kidney. Renal tissues were severely damaged and showed a large area of fibrosis. Besides, left ventricular hypertrophy and myocardial fibrosis were also observed. Conclusions AHP/ HL rat model is successfully constructed by feeding SHRs with high-fat diet for 23 weeks. Most importantly, this model exhibits progressive renal and cardiac alterations, similar to those of patients with HP/ HL. This HP/ HL rat model may become widely used for evaluation of HP/ HL therapeutic drugs.

Abstract:Objective During zebrafish embryo gastrulation, the embryonic body plan is established by coordinated movements, including epiboly, involution, and convergence and extension (CE) movements. The aim of this study was to examine the expression patterns and the developmental functions of isthmin 1 ( ism1) during zebrafish gastrulation. Methods First, the expression of ism1 was examined using whole mount in situ hybridization during zebrafish embryonic development. Next, antisense morpholino oligonucleotide against ism1 and ism1 mRNA were injected into zebrafish embryos, respectively, to investigate its role in CE movements. Results ism1 is an early zygotic gene that is expressed in the dorsal side at 30% epiboly stage and in the blastoderm margin with the highest level in the dorsal organizer at shield stage as revealed by whole-mount in situ hybridization experiments. Compared with the wild-type embryos, either gain- or loss-of-function of ism1 leads to severe defects of CE movements. Conclusions Ism1 plays an essential role in CE movements during zebrafish embryo gastrulation.

Abstract:Objective To investigate the induction of B?cell specific phenotype in classical Hodgkin lymphoma (cHL) upon all?trans retinoic acid (ATRA) incubation. Methods To construct B-cell specific promoter ( CD19, CD79a, CD79b) driven reporter plasmid with NEO cassette to realize stable transfection and selection of cHL reporter cells. To verify the intact integration by amplification of the promoter and luciferase sequences, and to functionally validate the B-cell specific promoter by ABF1 interference and luciferase assay. Repoter cells were incubated with various doses of ATRA and luciferase activity was detected at 24, 48 and 72 hours. Reporter cells were treated alone or in combination with 5-Aza and ATRA followed by luciferase assay. Endogenous B-cell specific genes (CD19, CD20, CD79a and CD79b) transcription and expression levels were detected by real-time PCR and immunoblot, respectively. The expression level of CD30 antigen on Hodgkin lymphoma cell membrane upon ATRA was assessed by flow cytometry. Results ATRA treatment stimulated B-cell specific signature in cHL cells including CD19, CD79a and CD79b while down-regulated their CD30 expression. Conclusions ATRA induces B-cell phenotype deficient cHL cells to regain their B-cell transcriptional program while abolishes their Hodgkin-specific machinery.

Abstract:Objective To observe the effect of platelet rich plasma (PRP)combined with core decompression on the steroid?induced avascular necrosis of the femoral head (SANFH) and on MMP/ TIM in rabbits. Methods < /b> A total of 42 New Zealand white rabbits were used in this study, and were randomly divided into 3 groups (decompression, combination and control groups, each n =14). The rabbit model of SANFH was established by i. m. injecting prednisolone acetate in the decompression group and combination group. The improved Landesberg method was used to make the platelet rich plasma. The decompression group received core decompression treatment while the combination group received PRP combined with core decompression for bone repair. X?ray photography of the hip joint of the two groups were taken at 2, 6 and 10 weeks after the surgery, and the Lane?Sandhu X?ray scores and new bone area ratio were compared. Venous blood samples of the 3 groups were collected and the bilateral femoral heads were taken for further examination. The left femoral heads were used for histopathological observation and the right ones were used to determine the expression of the mRNA of MMP?2,MMP?9, TIMP?1, and TIMP?2. Results The levels of MMP?2 and MMP?9 in the decompression group were higher than that in the combination group and control group after surgery. The levels of TIMP?1 and TIMP?2 in the decompression group were significantly lower than the combination group and control group (P < 0.05). The IL?6 level and the rate of empty bone lacunae in the decompression group were significantly higher than the combination group and control group (P < 0.05), and that of the combination group was higher than the control group (P < 0. 05). The combination group had a better joint imaging and histopathological evaluation than the decompression group after surgery. Conclusions Our findings demonstrate that PRP combined with core decompression can exert a positive effect on the MMP/ TIMP and bone tissue repair in the treatment of steroid?induced avascular necrosis of the femoral head in rabbits.

Abstract:Objective To explore the effects of Toddalia asiatica extract (TAE) on inflammatory cytokines in rats with myocardial ischemia and hyperlipidemia. Methods The early changes of hyperlipidemia were caused by feeding high fat diet for 4 weeks. Fifty male SD rats were randomly divided into 5 groups: the control group, model group, simvastatin group (3.5 mg/ kg), high-dose TAE group (280 mg/ kg) and low-dose TAE group (70 mg/ kg). At the end of the 4-weeks treatment, isoprenaline hydrochloride (Iso) was subcutaneous injected once a day for consecutive three days. The electrocardiogram (EKG),the heart,liver and fat indexes, the serum levels of TC, TG, HDL, LDL, TNF-α, INF-γ, IL-6 and IL-10, and the pathological changes of myocardium were examined. Results TAE significantly improved the Isoinduced EKG changes in rats, reduced the heart, liver and fat indexes,and decreased the levels of TC, TG, LDL, TNF-α, INF-γ and IL-6 and increased the contents of HDL and IL-10 (P < 0.05 for all), and improved the pathological damages of myocardium. Conclusions The Miao nationality medicine Toddalia asiatica extract may play an important protective role in cardiovascular diseases by regulating the balance of antiinflammatory and proinflammatory cytokines.

Abstract:Objective The effect of different doses of ethylnitrosourea (ENU) and cyclophosphamide (CP) on the loss rate of CD59 on peripheral blood erythrocytes was explored to optimize the detection method of Pig-a gene mutation. Methods According to the weight and loss rate of CD59 on peripheral blood erythrocytes, rats were divided into 4 groups: the control group, CP 40 mg/ kg group, ENU 10 mg/ kg group and ENU 40 mg/ kg group (n =6). The control group was injected i. p. with PBS, other groups were injected i. p. with corresponding solutions. The body weight of rats on days 0, 7, 14, 21, 28, 42 and 56 were recorded. At the same time, blood samples were collected and incubated with antibodies, and the loss rate of RBC CD59- was detected by flow cytometry. Results Compared with the control group, at different time points, the body weight and weight gain of ENU 10 mg/ kg group and ENU 40 mg/ kg group had no statistically significant difference (P > 0.05), while those in the CP 40 mg/ kg group were significantly decreased (P <0.05). The loss rate of RBC CD59- was significantly increased in the CP 40 mg/ kg group at 28, 42 and 56 days, ENU 10 mg/ kg group at 42 and 56 days, and ENU 40 mg/ kg group at 7, 14, 21, 28, 42 and 56 days, (P < 0.05). The results showed a dose-response relationship. Conclusions Under the conditions of this Pig-a mutation detection method , ENU is superior to CP on raising loss rate of RBC CD59- , ENU 40 mg/ kg is better than 10 mg/ kg, and 28 days is suitable as the test period.

Abstract:Objective To study the effect of autologous periosteum on the healing of tendon-bone interface in rabbit rotator cuff tear. Methods Supraspinatus tenotomy was performed on one side shoulder in 60 New Zealand white rabbits to establish the model of rotator cuff tear. The rabbits were randomly divided into two groups: the study group (used autologous periosteum to promote the suture fixation) and control group (simple suture fixation). The rabbits were sacrificed at 4, 8 and 12 weeks postoperatively with twenty rabbits sacrificed each time. Tissue samples of the tendon-bone interface was taken for histological examination and biomechanical test was performed to assess the strength of tendon-bone junction. Results At 4 weeks the study group showed extensive inflammatory cell infiltration, and a small amount of chondrocytes and extracellular matrix. At 8 weeks, the study group showed a large amount of immature chondrocytes arranged rather regularly. At 12 weeks, the tendon bone junction appeared similar to normal. In the study group, both the tendon-bone connection and the arrangement of chondrocytes were significantly better than the control group. The result of biomechanical testing showed that the highest tendon load in the study group was significantly higher than the control group (P < 0.05). Conclusions The use of autologous periosteum as a patch to strengthen the repair of the rotator cuff tear can effectively promote the healing of tendon bone interface, shorten the rotator cuff healing time and has good biological properties. This method provides an experimental basis for clinical rotator cuff repair surgery.

Abstract:Objective BALB/ c mutant curly mice and normal BALB/ c mice were genetically detected by microsatellite DNA marker analysis to detect the differential microsatellite loci between BALB/ c mutant curly mice and normal mice. Methods 38 microsatellite DNA loci were selected and their variation in the BALB/ c mutant curly mice, BALB/ c mutant hairless mice and normal BALB/ c mice were detected by multiplex fluorescence PCR and STR scanning genotyping. Results There were 27 the same microsatellite loci between the 38 microsatellite loci in BALB/ c mutant curly mice and normal mice, and there were 11 differential loci, with a mutation rate of 28. 9% (11/38). There were 30 the same sites between BABL/ c mutant hairless mice and normal mice, and there were 8 different loci, with a mutation rate of 21.1% (8/38). There were also 12 differential loci between BABL/ c mutant curly mice and hairless mice. Conclusions BALB/ c mutant curly mice have a higher mutation rate and are significantly higher than those of hairless mice, demonstrating that the mutations in curly mice and hairless mice are two completely different mutations. These results provide reliable theoretical data for the future study and development of BALB/ c mutant curly mice.

Abstract:Objective The ammonia level in air and heavy metals in drinking water were explored in a cynomolgus monkey feedlot. Methods Air ammonia from different communities and feeding patterns of cynomolgus monkeys were collected at three time-points per day and determined by Nessler’ s reagent spectrophotometry. Inductively coupled plasma mass spectrometry was applied to detect the level of heavy metals in drinking water from 5 different sampling sites in the feedlot, incluing the outlet of underground water, the water tank, both monkey cages equipped with PVC or iron pipes and sewage lagoon, respectively. Results Air ammonia levels in quarantine inspection flock cages (0.59 ± 0.03 mg/ m3 )were significantly higher than in the cages of both reproductive flock (0.34±0.03 mg/ m3 ) and sale flock (0.27±0.04 mg/ m3 ). The ammonia level in air in different feeding patterns ranks as following: cage rearing of quarantine inspection flock (0.59± 0.03 mg/ m3 ) > cage rearing of reproductive flock (0.48±0.02 mg/ m3 ) > captive bleeding of sale flock (0.30±0.02 mg/m3 ) > cage rearing of sale flock (0.25±0.01 mg/ m3 ) > captive bleeding of reproductive flock (0.22±0.02 mg/ m3 ). The air ammonia concentrations of both former flocks were statistically higher than the latter three flocks. The highest air ammonia level among different flocks and feeding patterns occurred in the morning, before waste discharge clean-up. The iron concentration in drinking water in the cages equipped with iron pipes was higher than Chinese drinking water standard. Conclusions The air ammonia level was lower than the Chinese air quality standard. The iron concentration in drinking water in the cages equipped with iron pipes was higher than the Chinese drinking water standard.

Abstract:Objective To Establish a loop-mediated isothermal amplification (LAMP) assay for detection of Salmonella in fecal samples of tree shrews, and report the result of preliminary application of this method. Methods LAMP primers were designed and synthesized according to the conserved sequence of Salmonella specific gene invA (invasive protein gene A). To optimize the reaction time and temperature by setting 10 reaction times (24 to 42 min) and temperature (57℃ to 66℃) and tested its specificity and sensitivity. At the same time, a conventional PCR test was performed to verify and compare with the LAMP assay. 91 fecal samples of wild-derived tree shrews were detected by the LAMP assay. Results The experimental condition was confirmed as 62℃ and 34 min. The sensitivity of Salmonella was 3.36 ×101 CFU/ mL, which was 10 to 100 times higher than that of conventional PCR assay. In 10 kinds of intestinal bacteria for LAMP amplification, Salmonella enteritidis and Salmonella paratyphi B were positive, the others were negative. Among the 91 samples of tree shrew fecal samples detected by the LAMP assay, the positive detection rate was 20.88%. The LAMP assay can be completed within 40 min, the result can be observed and judged visually by color changes. Conclusions The LAMP assay established in this study is convenient, rapid, sensitive and specific. It can be used as a rapid measure for large?scale detection of Salmonella in feces of tree shrews.

Abstract:A laboratory animal expert consultation system provides an interactive platform for industry experts and users. Based on intelligent mobile terminal used as the application supporter, the platform combines the internet with laboratory animal to share the experts’ knowledge, breaks through temporal and spatial restrictions, and resolves other problems such as poor timeliness of inquire service, knowledge data staticization, and irrational exploitation of internet to gain prompt inquire service. The platform would meet the needs of industry users for real?time aids and bridge the gap between industry experts and users. Thus, the transfer of expert’ knowledge is realized in pace with the increasing knowledge resource of this platform to accelerate the transformation of theoretical knowledge into the industry. Our study focuses on the system characteristics, the main frame structure, the design of function module and the system implementation.

Abstract:Long non-coding RNA (LncRNA) has attracted increasing attention over the recent years. It is closely related to the occurrence and development of human diseases based on numerous academic researches. Analysis of the biological function of LncRNA in depth may lead to great breakthroughs in clarifying pathogenesis of many diseases and provide possible targets for intervention. Recent studies have revealed the importance of LncRNA in the progression of heart failure (HF), the final stage of many cardiovascular diseases. This review summarized the new findings of LncRNA in prognosis and treatment of HF.

Abstract:The proliferation of tumor cells is regulated by a complex array of signaling pathways, among these signaling pathways, the programmed cell death. Autophagy and apoptosis are two types of programmed death. There are significant differences in their morphological and functional features, but they also have many links. Both apoptosis and autophagy are involved in activation, expression and regulation of a series of genes. By reviewing the research progress in recent years, this article will discuss the cellular regulation and molecular mechanisms of their related genes. Through summarizing the relationship between autophagy and apoptosis, it aims to get a better understanding of the mechanisms of autophagy and apoptosis in tumor progression, and looking at the perspectives for studies on the autophagy and apoptosis in tumor treatment.

Abstract:Breast cancer is the most common spontaneous malignancy in women, causing a serious threat to women’s health. Experimental animal models have been important research tools in bringing insights into the related mechanisms of breast cancer, as well as in delivering improved therapies for the disease. To reveal the pathogenesis of various breast cancers and facilitate the development of new drugs, ideal animal models of breast cancer should share some common tumor molecular characteristics and biological behavior with that of human breast cancer. In this review, five types of experimental animal breast cancer models were included, which may provide a basis for research on diagnosis and treatment of human breast cancer.

Abstract:Objective A state quo survey of laboratory animal resource in Guangdong province is performed to provide reference data for government management decision-making and market assessment of laboratory animals. Methods We used questionnaires focusing on the laboratory animal facilities with authorization by Guangdong Province Government, which mainly included the production and use of laboratory animals, the qualification of employees and the facilities space. Results The total production and use of laboratory animals (except for the eggs) had been increasing in the last four years. 1.57 million laboratory animals were produced and 0.754 million laboratory animals were used in 2016. There were 2352 employees, roughly the same as in 2015. The facilities space for breeding was 121008 m2 , and for animal experiment was 73470 m2 , which were rising in the past three years. Conclusions In order to reinforce the industry development of laboratory animals in Guangdong province, some suggestions were given in our study, such as facilitating the application of superiority resource including non-human primate and aquatic laboratory animals, supporting the standardization production of several scarce mice and rats, improving relevant employees’ overall level and constructing laboratory animal facilities sharing platform.

Abstract:Laboratory red crucian carp is a native fish in China. It has been an established inbred fish strain, the laboratory red crucian carp C1HD strain. Based on the standardization research and verification test of laboratory red crucian carp, the local standard of Hunan Province, the called “Laboratory Fish Genetic Quality Control of Laboratory Red Crucian Carp C1HD Strain”, has been established. This standard includes preface, scope, normative reference documents, terms and definitions, major biological characteristics, mating and breeding method , breeding environment,genetic quality testing and appendix A, B, C and so on.
